摘 要:目的研究烟曲霉对巨噬细胞吞噬能力及细胞活性的影响规律。方法通过免疫荧光法测定巨噬细胞吞噬烟曲霉后吞噬量的变化,同时利用MTT比色法测定巨噬细胞的细胞活性变化。结果在吞噬初期(20min),巨噬细胞吞噬乳胶颗粒的速度明显高于吞噬孢子(P<0.05),然而最终(70min)吞噬两种颗粒的能力无明显差异(P>0.05);巨噬细胞吞噬休眠孢子及热灭活孢子的量无显著差异(P>0.05);另外,休眠孢子、热灭活休眠孢子及膨胀孢子均可显著降低巨噬细胞的细胞活性(均P<0.05),而烟曲霉菌丝和乳胶颗粒对巨噬细胞的细胞活性无明显影响(P>0.05)。无色素的烟曲霉白化株休眠孢子不能使巨噬细胞活性降低(P>0.05)。结论烟曲霉孢子表面色素可能是降低巨噬细胞活性的影响因素。Objective To study the effect of different live-forms of Aspergillus fumigatus on both macrophage phagocytosis and cytoactivity.Methods Phagocytosis of Aspergillus fumigatus conidia and latex beads by macrophage was determined by GFP-expressing conidia and immunofluorescence;macrophage cytoactivity was measured by microplate-MTT colorimetric assay.Results At the early phase of phagocytosis(around first 20 min),latex beads were phagocytized more rapidly than conidia by macrophage (P〈0.05),however,there was no significant difference in phagocytosis capacity between two of them(P〉0.05).Moreover,heat-killing pretreatment on conidia did not affect its phagocytosis by macrophage (P〉0.05 ).In addition,cytoactivity of macrophage was inhibited by the interaciton of macrophage with live rest conidia,swollen conidia and heat-killed conidia(P〈0.05),but not hyphae and latex beads (P〉0.05).Albino Aspergillus fumigatus conidia did not inhibited the cytoactivity of macrophage(P〈0.05).Conclusion Melanin,which is present on the surfaces of Aspergillus fumigatus conidia,may be able to affect the cytoactivity of macrophage.
