摘  要:目的:通过观察血管内皮生长因子(VEGF)、骨形态发生蛋白-2(BMP-2)在骨折愈合过程中的表达及分布情况,探讨骨折一号方对实验性骨折愈合过程中VEGF、BMP-2表达的影响和促进骨折愈合的机制。方法:制作60只兔桡骨骨折模型,随机分为空白组、生理盐水对照组及骨折一号方组,术后1d骨折一号方组即喂服试药,对照组喂服生理盐水。分别于术后第3、6、9、14天取样,进行组织学切片、VEGF、BMP-2免疫组化染色,并采用图像分析系统对VEGF、BMP-2的表达进行分析。结果:①骨折后断端均有VEGF表达。术后第3、9天出现两个峰值。实验组较对照组各时相点VEGF表达量均明显增高。②骨折后断端均有BMP-2表达,各时相点未出现峰值,而是呈两条近乎平行线,随时间的推移而呈增高趋势。在各时相点,骨折一号方组BMP-2表达均高于对照组和空白组(P<0.05)。结论:骨折一号方能显著增高各时相点VEGF、BMP-2表达量,使骨折断端血循环重建及成骨过程加快,这可能是其产生临床疗效的机制之一。Objective:To explore the effect of Guzheyihaofang on experimental fracture healing VEGF, BMP-2 expressions and the mechanism of fracture healing by observing the vascular endothelial growth factor (VEGF)and bone morphogenetic protein -2 (BMP-2) in fracture healing expression and distribution. Methods: 60 of bone fracture were randomly divided into model group, normal saline control group and Guzheyihaofang group. After 3, 6,9,14 days, through sampling, histological sections, VEGF, BMP-2 immunohistochemical staining and image analysis system, VEGF, BMP-2 expression were analyzed in both groups. Results: ① There were VEGF expressions after the fracture ends and two peaks. In the experimental group than in the control group at each time point VEGF expressions were significantly increased.②There were BMP-2 expressions in the fracture ends. It was observed that the peak time point gave way to two parallel lines, with rising tendency. At each time point, the expression of BMP-2 in Guzheyihaofang group were higher than in the other groups (P<0.01),with significant differences. Conclusion: Guzheyihaofang can increase significantly at all time points VEGF and BMP-2 expressions, and facilitate the reconstruction of fracture fragments and the blood circulation and the process of bone formation.
