摘 要:建立灵敏、准确地测定人血浆中水杨酸浓度的HPLC法。样品经过酸化处理后,以甲苯-正辛醇(4:6,V/V)为萃取溶剂、NaOH水溶液(pH=10)为接收相进行中空纤维膜微萃取。在该条件下,血浆中水杨酸浓度在2-250 ng·mL^-1范围内,线性关系良好(r=0.9994);检测限为0.7ng·mL^-1(S/N=3);精密度(RSD)〈8%(n=5);回收率为87.6%-102.0%。该方法灵敏、有效、所需有机溶剂少、富集效率高,适于血浆中水杨酸浓度的测定。A sensitive,accurate method to determine salicylic acid in human plasma by HPLC was developed.Using hollow fiber liquid phase microextraction as a sample preparation technique,the optimum experimental conditions were the following: binary-solvent of toluene:n-octanol(4∶6,V/V) as extraction solvent,HCl solution as donor phase,NaOH solution at pH 10 as acceptor phase.Under the optimum conditions,the linear range of salicylic acid was 2-250 ng·mL^-1(r=0.9994),the limit of detection was 0.7 ng·mL^-1,relative recoveries were 87.6%-102.0% with RSD less than 8%.The method was sensitive and effective for determination of salicylic acid in human plasma that required little organic solvent and provided a high degree of enrichment.
