清热解毒中药对流感病毒FM_1株感染所致小鼠肺组织病理损伤的影响

摘　　要：目的动态观察清热解毒中药对流感病毒感染所致小鼠肺组织病理损伤的影响。方法将96只健康雄性BALB/cAnN小鼠随机分为空白对照组、流感病毒感染模型组、中药治疗组,每组按照流感病毒感染1、3、5、7天不同时相再分为4个小组,每小组8只小鼠。用流感病毒亚洲甲型鼠肺适应株(FM1株)感染小鼠建立流感病毒性肺炎模型。在FM1株感染不同时相,中药治疗组用清热解毒中药混悬液(由生石膏30g、黄芩10g、生甘草3g组成,相当于成人临床剂量5.58g.kg-1.d-1)灌胃,空白对照组及流感病毒感染模型组用等量生理盐水灌胃。在FM1株感染1、3、5、7天,摘眼球放血处死小鼠,打开胸腔摘出全肺,分别用肉眼、光学显微镜、透射电镜动态对比观察肺组织的病理学变化。结果肉眼观察,中药治疗组小鼠肺脏病变在FM1株感染后3～7天,较病毒感染模型组明显减轻,仅有少量渗出,病灶范围较小,病变数量较少。光镜下观察,中药治疗组在FM1株感染后3～7天,肺组织病变程度较病毒感染模型组明显减轻,肺组织病灶较小,炎症细胞分布密度较低,细支气管上皮脱落减轻,支气管管腔内炎症细胞渗出明显减少,小血管内皮细胞肿胀减轻。电镜下观察,中药治疗组小鼠肺组织病变较病毒感染模型组明显减轻,肺泡结构破坏减轻,可见Ⅱ型细胞增生,肺内毛细血管内皮细胞形态大致正常,无肿胀。结论清热解毒中药能够显著改善FM1株感染所致小鼠肺脏的炎性病变,对FM1株感染所致小鼠肺组织病理损伤具有较强的修复作用。Objective To observe dynamically the effect of heat-clearing and detoxicating herbs on the pathological injury of lung tissue in mice infected with influenza virus FM1.Methods Ninety-six healthy male BALB/cAnN mice were randomly divided into three group：normal control group,influenza infection model group and Chinese medicine treatment group.Each group was divided into 4 subgroups according to different time points（1,3,5 and 7 days after mice were infected with influenza virus）,8 mice in each subgroup.The model was made with the mice infected with FM1 influenza.During each phase of the infection,the mice in Chinese medicine treatment group were administered intragastrically with suspension of heat-clearing and detoxicating Herbs（Gypsum 30 g,Scutellaria 10 g,raw licorice 3 g,equivalent to adult clinical dose 5.58 g·kg-1·d-1）,meanwhile the mice in normal control group and influenza infection model group were administered intragastrically with the same amount of normal saline.On the 1st、3rd、5th and 7th day of infection,the mice were sacrificed and the lungs were take out.Then the dynamically pathological changes of lung tissue were observed with naked eyes,optical microscope and transmission electron microscopy respectively.Results According to the observation with naked eyes,the histological changes of lung in Chinese medicine treatment group were significantly slighter than those in influenza infection model group on day 3 and day 7,with a smaller amount of leakage,smaller range of focus and less lesions.According to the observation with optical microscope,the histological changes of lung in Chinese medicine treatment group were significantly alleviated in samller lung lesions,lower inflammatory cells density,less bronchioles epithelium loss,less inflammatory cells effusion in bronchial lumen and slighter endothelial cells of small blood vessels swelling.According to the observation with transmission electron microscopy,the histological changes of lung in Chinese medicine treatment group were signi
