摘  要:目的观察脂多糖致急性肺损伤大鼠肺泡上皮细胞Occludin和Zo-1蛋白表达的变化。方法将30只SD雄性大鼠随机分为正常对照组和脂多糖致伤组。气管切开后滴注脂多糖0.5mL/kg(0.2mg/mL)复制急性肺损伤模型,分别于伤后4h和24h取材,采用免疫组化染色法测定肺组织Occludin和Zo-1蛋白的表达。结果脂多糖致急性肺损伤后4h肺泡上皮细胞Occludin和Zo-1蛋白表达明显下降(P<0.05),24h后有所恢复,但仍比对照组表达减少。结论急性肺损伤早期即出现紧密连接蛋白表达减少,Occludin和Zo-1表达与急性肺损伤发生密切相关,急性肺损伤后紧密连接蛋白表达与肺水肿的发生和发展密切相关。Objective To observe the changes of expression of Occludin and Zo-1 on alveolar epithelial cells in LPS induced lung injury in rata. Methods A total of 30 SD rats were randomly assigned to the control group (n = 10) and the injured group(n = 20) in which the lung injury was induced by intratraeheal instillation of LPS at dose of 0.5mL/kg. The expression of Zo-1 and Occludin on the alveolar epithelial cells were examined at 4h, 24h after injury (n = 10 at each time point). Results After lung injury, there was significant decrease of the expression of Zo-1 and 0ccludin on alveolar epithelial cells. On 4h after injury when all changes were most significant, as compared with control group. On 24h after injury the changes of the expression of Zo-1 and Occludin improved but still lower than those of eontrel group. Conclusion The level of tight junction decreases in early acute lung injury, and the levels of Zo-1 and Occludin decrease in the early stage of acute lung injury. The expressions of Zo-1 and Occludin on the alveolar epithelial cells are associated with acute lung injury.
