摘 要:目的:研究核糖体DNA(rDNA)的ITS序列在胡椒属大叶蒟及其近缘药用植物及药材鉴定中的适用性。方法:聚合酶链反应(Polymerase Chain Reaction,PCR)扩增ITS序列测序,用DNAssist version2.2软件进行序列比对,并计算同源性。结果:胡椒属药材ITS序列与外类群ITS序列比对同源性均小于67.0%,属内序列间两两比对同源性均大于82.0%,同种植物则大于99.0%。结论:ITS序列作为胡椒属药材鉴定依据具有一定的适用性和可靠性。Objective: Study on the applicability of rDNA ITS sequence in identification of medicinal plants from Piper L.and its related species.Methods: The ITS regions of these species were amplified by PCR and sequenced,and their sequences were analyzed by DN Assist Version 2.2.Results:Homologous alignment indicated that the consanguinity of the ITS sequences between Genus Piper and outgroups was lower than 67.0%,and that within genus Piper was higher than 82.0%.For the same species,the consanguinity was higher than 99.0%.Conclusions: The ITS sequences may serve as reliable molecular markers for identification of Genus Piper and its related species.
