摘 要:目的:观察亚甲蓝溶液对退变椎间盘动物模型NO浓度的影响,进而探讨亚甲蓝注射治疗椎间盘源性腰痛的可能机制。方法:实验选用10只新西兰大白兔通过纤维环穿刺法建立L4/5和L6/7椎间盘退变动物模型,经MR I检查证实椎间盘退变后在L5/6和L6/7椎间盘内注入亚甲蓝溶液,L4/5椎间盘作为阳性对照,L5/6椎间盘作为阴性对照,L6/7椎间盘作为实验组椎间盘。饲养2周后将动物处死,取下椎间盘标本并对标本行组织学染色观察,同时应用NO试剂盒检测椎间盘标本中NO浓度的变化。结果:实验组椎间盘标本中的NO浓度较阳性对照组标本中的浓度明显减低,经统计学检验,其差别有统计学意义。结论:亚甲蓝能够抑制退变椎间盘内NO的产生,这可能是亚甲蓝注射治疗椎间盘源性腰痛的可能机制之一;亚甲蓝溶液对正常椎间盘是安全的,不引起椎间盘退变。Objective: To observe the influence of methylene blue (MB) on the NO concentration in degenerative intervertebral disc, and furtherly probe the possible mechanism of MB injection in therapy discogenic low back pain. Methods: We establish animal model of disc degeneration by puncture the intervertebral disc annulus fibrosus, when the discs get degenerated, MB is injected into the degenerative discs, then we observe the changes of concentration of inflammatory mediators by the method of Histology staining and Molecular Biology. What is more, we conduct statistical tests to related records. Conclusion: The abnormal inflammation of the degenerative disc can be forbidden by the injected MB. So we can conclude that which may be one of the possible mechanisms for the therapy of Methylene blue injected to cure Discogenic low back pain. What is more, we also found that MB is safety to the normal disc and may not start the process of disc degeneration during this study.
