摘  要:目的 研究人肝脏星状细胞( HSC) 上糖基化终产物受体(RAGE) 的表达。方法 用生物素将体外制备的糖基化终产物( AGEs - BSA) 标记成配基,以细胞化学法显示细胞表面受体高亲和性结合。用反转录聚合酶链反应( RT-PCR) 检测AGEs 受体基因的表达。结果 细胞化学法显示AGEs - BSA 与HSC 呈高亲和性结合,部分被内化。用按人RAGE 基因序列设计的引物,反转录扩增出283 bp DNA 片段。结论 人肝脏星状细胞表达的AGEsObjective To investigate expression of receptor for advanced glycosylation end products (AGEs) in cultured human hepatic stellate cells (HSC). Methods AGEs-BSA prepared in vitro was labeled with biotin to demonstrate binding of AGEs to receptor for AGEs (RAGE). High affinity binding of cell surface was shown with cell chemistry method. RT-PCR was performed with oligonuclotide primers according to human RAGE gene sequence to detect mRNA expression of RAGE. Results High affinity binding of biotin labeled AGEs-BSA to surface of cultured HSC was identified. 283 bp DNA was amplified which was consistent with predicted fragment according human RAGE sequence. Conclusion The results suggested that expression of RAGE in HSC might be of significance in pathogenesis of accelerated hepatic fibrosis in diabetic patients.
