摘  要:目的探讨白藜芦醇(Res)诱导人皮肤癌细胞系(A431细胞)凋亡的可能信号分子,为临床相关疾病的治疗提供新的实验依据。方法培养的A431细胞分为:对照组(A组)、Res(B组)、Res加SP600125(C组)以及单纯SP600125组(D组)。免疫荧光染色、Western blot及相差显微镜观察等方法检测细胞形态和caspase-3、BAX、BCL-2、JNK以及P-JNK蛋白分布和含量。结果 Res处理后,A431细胞出现较典型的细胞凋亡形态特征,同时可见BAX/BCL-2比值升高,caspase-3蛋白表达增加,磷酸化(P-JNK蛋白)明显增加,且集聚在细胞核(由225 463±672增至289 469±3 729,P<0.05);经SP600125预处理后,上述改变明显减少(降至248 198±4 367,P<0.05),A431细胞的生长状态与对照组相似。结论 Res可能首先使JNK蛋白磷酸化,进入细胞核,发挥抑制BCL-2蛋白表达,上调BAX和caspase-3蛋白表达,进而引起细胞凋亡。Objective To investigate a potential signal molecule of resveratrol(Res)-induced apoptosis of human skin cancer cell line,A431 cells.Methods A431 cells were divided into 4 groups as group A(controls),group B(treated with Res),group C(pre-treated by SP600125) and group D(inhibitor control,SP600125 only).Phase contrast microscopy,Western blot and immunofluorescence staining were used to observe morphology and the expression of caspase-3,BCL-2,BAX,JNK and P-JNK in A431 cells.Results Apoptosis was often found after Res treatment in the A431 cells.Meanwhile,the expression of caspase-3 and rate of BAX/BCL-2 increased,P-JNK expression increased significantly and aggregated in the nucleus(increased to 289 469±3 729 from 225 463±672,P0.05).Pre-treated with SP600125,the above changes decreased significantly(248 198±4 367,P0.05),and the growth of A431 cells was not different from that of controls.Conclusion The phosphorylated JNK entering into the nucleus to regulates the expression of BCL-2,BAX and caspase-3 are involved in the signal molecule of Res-induced apoptosis in A431 cells.
