微囊化大鼠卵巢细胞对去卵巢大鼠骨代谢的影响

摘  要:目的:观察去卵巢大鼠微囊化卵巢细胞移植后不同时期的骨代谢指标的变化,探讨微囊化卵巢细胞作为防治绝经后妇女骨质疏松方法的可行性。方法:将12周龄SD雌性大鼠36只随机分成假手术组(SHAM组)、去卵巢组(OVX组)和微囊化卵巢细胞移植组(MOVT组)。术后10天MOVT组大鼠腹腔注射微囊化卵巢细胞2 ml,SHAM组和OVX组注射0.9%NaCl溶液2 ml,观察12周;术后4、8、12周尾静脉采血测定血清E2及骨代谢指标ALP、TRAP、OC、HYP的浓度或活性。结果:与SHAM组大鼠比较,OVX组大鼠血清E2在术后4周即有非常显著的下降(P<0.01),且随着时间延长而持续下降;OVX组大鼠血清TRAP、HYP、ALP、OC水平在术后均显著升高,其中TRAP、HYP在术后8周达到最高峰(P<0.01);而ALP、OC则在术后12周达到最高峰(P<0.01)。与OVX组大鼠比较,MOVT组大鼠血清中E2水平在移植后4周显著升高(P<0.05),移植后8周、12周差异显著(P<0.01);血清TRAP和HYP水平在移植后4周即明显降低(P<0.05),到移植后8周差异有显著性意义(P<0.01);而ALP和OC水平在移植后8周开始降低,其中ALP更加显著(P<0.05),到移植后12周达到最小值(P<0.01)。结论:微囊化卵巢细胞移植具有预防去卵巢大鼠骨质疏松发生的作用,微囊化卵巢细胞移植作为临床防治绝经后骨质疏松的一种手段具有良好的应用前景。Objective:To observe the changes of indexes of bone metabolism at different periods after transplantation of microencapsulated ovarian cells of ovariectomized rats,explore the feasibility of microencapsulated ovarian cells as methods to prevent and treat osteoporosis of menopausal women.Methods:36 female SD rats aged 12 weeks old were divided into sham operation group(SHAM group),ovariectomized group(OVX group) and transplantation group of microencapsulated ovarian cells(MOVT group).At 10 days after operation,the rats in MOVT group were treated with intraperitoneal injection of microencapsulated ovarian cells(2 ml),the rats in SHAM group and OVX group were treated with injection of 0.9% NaCl solution(2 ml),then the rats were followed up for 12 weeks;at 4,8 and 12 weeks after operation,blood samples were obtained from caudal vein to detect the level of serum estradiol and the contents of indexes of bone metabolism(including ALP,TRAP,OC and HYP).Results:Compared with SHAM group,the level of serum estradiol at 4 weeks after operation in OVX group decreased significantly(P0.01),decreasing with time;after operation,the serum levels of TRAP,HYP,ALP and OC in OVX group increased significantly,and the serum levels of TRAP and HYP peaked at 8 weeks after operation(P0.01),while the serum levels of ALP and OC peaked at 12 weeks after operation(P0.01).Compared with OVX group,the level of serum estradiol at 4 weeks after operation in MOVT group increased significantly(P0.05),there was significant difference in the level of serum estradiol between 8 weeks after operation and 12 weeks after operation(P0.01);the serum levels of TRAP and HYP at 4 weeks after operation decreased significantly(P0.05),there was significant difference at 8 weeks after operation(P0.01);the serum levels of ALP and OC decreased at 8 weeks after operation(P0.05) and peaked at 12 weeks after operation(P0.01).Conclusion:Transplantation of microencapsulated ovarian cells in ovariectomized r
