摘 要:目的:建立使用超高效液相色谱(UPLC)测定钩藤不同提取工艺样品中钩藤碱(Rhychophylline)的含量方法.方法:采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×50 mm,1.7μm),甲醇-5 mmol/L乙酸铵水溶液用氨水调pH=9.8(60∶40)流动相进行等度洗脱,检测波长为240 nm.结果:钩藤碱在0.026~0.520μg范围内有良好的线性关系(r=0.9999),平均回收率(n=6)为100.3%.结论:该方法简便、快速、准确可靠,可应用于钩藤药材中钩藤碱含量的测定.The amount of rhychophylline in different extraction of samples was determined by UPLC with waters Acquity UPLC BEH C18 column(2.1 mm × 50 mm,1.7 μm),methanol and 5 mmol / L ammonium acetate solution adjusted pH = 9.8 with ammonia-methanol as mobile phase(40∶ 60)i,socratic elutiont,he detection wavelength was 240 nm.The calibration curve is linear in the range of 0.026 ~ 0.520 μg with correlation coefficient of 0.9999.The average recovery of rhychophylline was 100.3%.The method is rapid,accurate,reproducible in determination rhychophylline in Ramulus Uncariae cum Uncis.
