New genotype of Sporothrix schenckii in Xinjiang

摘  要:目的:利用多聚酶链反应随机扩增多态性DNA(polymerasechain reaction-random amplified polymorphic DNA,PCRRAPD)法对新疆地区固定型和皮肤淋巴管型孢子丝菌病致病菌进行基因分型,并与北京和哈尔滨的菌株基因型比较,探讨新疆地区申克孢子丝菌的基因特点。方法:采用随机单核苷酸引物OPB 07、OPAA 11、OPD 18用RAPD法对来自新疆、北京和哈尔滨的18株申克孢子丝菌进行DNA多态性分析。结果:新疆的固定型孢子丝菌病致病菌有独特的基因型,新疆的淋巴管型孢子丝菌病分离菌株和北京、哈尔滨的菌株间无基因差异。结论:新疆地区申克孢子丝菌显示新的基因型。新疆和北京的固定型孢子丝菌病致病菌有种内基因差异,而新疆与北京、哈尔滨的淋巴管型孢子丝菌病的致病菌间无种内基因差异。引物OPB07和0PAA 11结合PCR-RAPD法是申克孢子丝菌基因分型的可行性方法之一。Objective:To study genotypes of Sporothrix schenckii strains isolated from lymphangitic and fixed types of sporotrichosis in Xinjiang by polymorphic chain reaction random amplification of polymorphic DNA(PCR-RAPD) method,and compare the genotypes of the strains from Beijing and Haerbin,to explore the genotypic characteristics of Sporothrix schenckii in Xinjiang. Methods:DNA polymorphism of 18 samples isolated from different clinical types of sporotrichosis from Xinjiang,Beijing and Haerbin were performed by PCR-RAPD method with random single nucleotide primers OPB07,OPAA11,and OP D18.Results; The strain from fixed type of sporotrichosis in Xinjiang showed unique genotype.There were no genetic variation found among the strains from lymphangitic sporotrichosis in Xinjiang,Beijing and Haerbin.Conclusions:Sporothrix schenckii in Xinjiang showed new genotype.There is intraspecies diversity between the causative agents of fixed type sporotrichosis in Xinjiang and Beijing, while no intraspecies diversity among the causative agents of lymphangitic sporotrichosis in Xinjiang,Beijing and Haerbin.PCRRAPD with primers OPB 07 and OPAA 11 is one of reliable methods for genotyping of Sporothrix schenckii.
