摘 要:目的通过检测初、复发翼状胬肉组织块分泌的相关因子并作为观察指标,了解汉防己甲素(Te-trandrine,Tet)对翼状胬肉病变的作用。方法收集初、复发翼状胬肉和正常球结膜组织块进行体外培养,并将初、复发翼状胬肉组织各分为无药组和药物干预组,后者加10-5 mol.L-1的Tet作用48 h。ELISA法检测上清液中血管内皮生长因子(Vascular endothelial growth Factor,VEGF)、白介素-1β(Interleukin-1β,IL-1β)的含量。结果 (1)初发翼状胬肉无药组的组织上清夜VEGF含量较初发翼状胬肉药物干预组及正常结膜组高(P<0.05),而初发翼状胬肉药物干预组与正常结膜组的VEGF含量相当(P>0.05)。复发翼状胬肉组织无药组的VEGF含量低于正常结膜组织。(2)初、复发翼状胬肉无药组上清液中IL-1β的含量与正常结膜组相比,差异无统计学意义(P>0.05)。结论 Tet对初发翼状胬肉组织中VEGF的分泌有抑制作用,而VEGF可能与初发翼状胬肉血管的形成及病情进展密切相关。VEGF在复发翼状胬肉中的作用及IL-1β在翼状胬肉中的作用尚需进一步研究。OBJECTIVE To examine the level of cytokine VEGF,IL-1β pterygium(primary,recurrent)tissue secreted in organ culture and observe the change of the cytokines in effect of tetrandrine(Tet).METHODS Pterygium(primary,recurrent)and conjunctival tissue were cultured in a serum-free medium respectively under the same experimental conditions.Primary and recurrent pterygium tissue samples were divided into non-drug group and Tet-treatment group respectively.The latter was cultured in a 10-5 mol·L-1 Tet medium for 48h.ELISA method was used to detect the concentration of vascular endothelial growth factor(VEGF)and interleukin-1β(IL-1β)in supernatants from organ culture.RESULTS(1)The level of VEGF was higher in medium from cultured primary pterygium non-drug group compared to primary pterygium Tet-treatment group and normal conjunctival tissue(P0.05).The level of VEGF in primary pterygium Tet-treatment group and cultured normal conjunctival tissue had no statistical difference.The level of VEGF was not higher in the medium from current pterygium tissue compared to normal conjunctival tissue.(2)There was no statistical difference in the level of IL-1β in the medium between cultured pterygium(primary,recurrent)non-drug group and normal conjunctival tissue.CONCLUSIONS Tet,10-5mol·L-1,dramatically decreased the amount of VEGF secreted by cultured primary pterygium tissue.VEGF maybe closely related with primary pterygia blood vessel formation and development.More research needs to be done to investigate the effect of VEGF on current pterygium and IL-1β on pterygium.
