摘  要:目的研究注射膜联蛋白5后雄性SD大鼠睾丸类固醇急性调节蛋白(Steroidogenic acute regulatory protein,StAR)、胆固醇侧链裂解酶(P450 cholesterol side-chain cleavage enzyme,P450scc)和3β-羟类固醇脱氢酶(3 beta-hydroxysteroid dehydrogenase,3β-HSD)表达的改变,初步探讨膜联蛋白5影响睾酮分泌的机制。方法将20只SD大鼠随机分为对照组和实验组,实验组又分为7.5μg/kg组、15μg/kg组和30μg/kg组,每组5只。对照组大鼠腹腔注射等量的pH8.0 Tris-HCl,实验组腹腔注射不同剂量的膜联蛋白5,1次/d,连续20 d。用RT-PCR方法分别检测对照组和各实验组SD大鼠睾丸组织中睾酮合成相关的StAR、P450scc和3β-HSD mRNA的表达变化,并用Western blot方法检测以上3种物质蛋白质水平的变化。结果与对照组相比,在mRNA水平上,7.5μg/kg组和15μg/kg组大鼠睾丸组织P450 scc mRNA表达分别增加了25.9%[(0.68±0.04)vs(0.54±0.03),P<0.01]和27.8%[(0.69±0.04)vs(0.54±0.03),P<0.01];3β-HSD mRNA表达分别升高了32.9%[(1.05±0.07)vs(0.79±0.10),P<0.01]和53.2%[(1.21±0.07)vs(0.79±0.10),P<0.01];而30μg/kg组表达差异无统计学意义;在蛋白水平上,15μg/kg组大鼠睾丸组织P450 scc蛋白表达提高了34.2%[(0.37±0.04)vs(0.28±0.02),P<0.01],7.5μg/kg组和15μg/kg组的3β-HSD表达也分别增加了14.7%[(1.53±0.10)vs(1.34±0.05),P<0.01]和24.4%[(1.67±0.14)vs(1.34±0.05),P<0.01];而StAR的表达无论在转录水平还是在蛋白水平上,较对照组差异均无统计学意义。结论膜联蛋白5可通过调节P450scc与3β-HSD的表达来调节睾酮合成。Objective The level of serum testosterone is increased after injection of annexin5 in SD rats.This study aimed to investigate the changes in the expressions of StAR,P450scc and 3β-HSD in the rat testis and discuss the mechanism of annexin 5 affecting testosterone synthesis.Methods Twenty experimental SD rats were intraperitoneally injected with annexin 5 at 7.5 μg/kg,15 μg/kg and 30 μg/kg once a day for 20 days.The control groups were injected with equal volume of pH 8.0 Tris-HCl.The expressions of StAR,P450scc and 3β-HSD at the mRNA and protein levels were detected by RT-PCR and Western blot,respectively.Results Compared with the control groups,the mRNA expression of P450 scc in the 7.5 μg/kg and 15 μg/kg groups increased 25.9%(0.54±0.03 vs 0.68±0.04,P0.01) and 27.8%(0.54±0.03 vs 0.69±0.04,P0.01),respectively,and that of 3β-HSD increased 32.9%(0.79±0.10 vs 1.05±0.07,P0.01) and 53.2%(0.79±0.10 vs 1.21±0.07,P0.01),but no significant difference was found in the 30 μg/kg group;the protein expression of P450 scc increased 34.2% in the 15 μg/kg group(0.28±0.02 vs 0.37±0.04,P0.01),and that of 3β-HSD increased 14.7% in the 7.5 μg/kg group(1.34±0.05 vs 1.53±0.10,P0.01) and 24.4% in the 15μg/kg group(1.34±0.05 vs 1.67±0.14,P0.01),respectively.The expression of StAR showed no statistically significant differences between the experimental and control groups either at the mRNA or protein level.Conclusion Annexin 5 regulates testosterone production by promoting the expressions of P450 scc and 3β-HSD.
