摘  要:[目的]观察亚慢性砷暴露对小鼠脑组织单胺类神经递质浓度的影响。[方法]昆明种小鼠70只,随机分为6组,即饮用水对照组、1 ppm As2 O3染砷组、2 ppm As2 O3染砷组、4 ppm As2 O3染砷组、2个保护组(4 ppm As2 O3+150 mg/kg牛磺酸和4 ppm As2O3+45 mg/kg维生素C)。除高剂量染毒组(4 ppmAs2O3组)20只外,其余每组10只小鼠。用高效液相色谱-荧光检测法(HPLC-Flu)测定单胺类神经递质5-羟色胺(5-HT)、多巴胺(DA)和去甲肾上腺素(NE)的浓度。[结果]与对照组比较,各染砷组小鼠脑中5-HT、DA和NE 3种单胺类神经递质浓度均明显降低(P<0.05),而2个保护组3种单胺类神经递质浓度,除牛磺酸组的5-HT外,均显著高于高剂量染砷组(P<0.05)。随染毒天数增加,高剂量染砷组小鼠脑中3种单胺类神经递质均逐渐降低,与染毒第0天比较差异有显著性意义(P<0.05)。[结论]亚慢性砷暴露能够降低小鼠脑中5-HT、DA和NE的浓度。牛磺酸和维生素C对砷导致NE、DA和5-HT水平降低,有一定的保护作用。[Objective] The present study was to elucidate whether low-dose exposure to arsenic would affect monoamine neurotransmitters levels in mice brain. [Methods] Seventy mice were randomly divided into six groups.The mice of Group 1 received drinking water alone as control.Those of Group 2,Group 3,Group 4,Group 5and Group 6 received 1 ppm,2 ppm,4 ppm As2O3,4 ppm As2O3+150 mg/kg taurine,and 4 ppm As2O3+45 mg/kg Vit C,respectively.Three monoamine neurotransmitters(norepinephrine,dopamine,serotonin) in cerebrum of the mice were measured by HPLC with fluorescence detector.[Results] Three monoamine neurotransmitters in cerebrum of the mice exposed to arsenic decreased significantly,compared to the control group.Oral administration of taurine and Vit C were found to be very effective in prevention for the brain injury of experimental mice.With the increasing of days exposed,the levels of three monoamine neurotransmitters were decreasing.[Conclusion] Subchronic low-dose exposure to arsenic can decrease the concentration of three monoamine neurotransmitters in brain.Taurine and Vit C may distinctively resist the injury of nervous system induced by arsenic.
