糖皮质激素对血清剥夺成人骨髓间充质干细胞及其RS细胞亚群的保护作用

摘  要:目的:探讨血清剥夺(SD)导致成人骨髓间充质干/祖细胞群(hMSCs)中成熟MSC(mMSC)和快速自我更新细胞(RS细胞)亚群凋亡、坏死及糖皮质激素(GC)的保护作用。方法:以无血清体外培养hMSCs为模型,经细胞形态学观察和Annexin V-PI双染流式细胞仪检测,观察细胞形态变化、核变化及活性/凋亡率变化。结果:SD 12h后hMSC皱缩变圆、染色质固缩,出现凋亡各阶段形态特征;与对照组相比mMSC及RS活性明显降低并且凋亡率升高(P<0.01),与血清剥夺组相比添加10nM DXM后细胞活性显著提高(P<0.01);并且RS细胞亚群对SD环境表现出更强的耐受性,与mMSC亚群相比存活率提高2.75倍,凋亡率降低近三分之一,并在糖皮质激素的保护作用下继续保持良好增殖。结论:糖皮质激素可以提高干细胞自我保护能力,并且RS细胞具有更强的自我保护能力和自我调节机制。Objective: To study the protective effect of glucocorticoid(GC) on mature MSC(mMSC) and rapid self-renewing cells(RS cells) in adult bone marrow mesenchymal stem / progenitor cell population(hMSCs).Methods: By cell morphology and Annexin V-PI double staining flow cytometry the changs were observed in cell morphology,nuclear changes and the activity / apoptosis rate changes.Results:hMSC shrinkage,chromatin condensation,morphological stages of apoptosis were observed.Compared with the control group,mMSC and RS activity was significantly decreased and apoptosis increased (P0.01);compared with serum deprived group,cell activity was significantly increased(P0.01) after adding 10nM DXM.The viability and the apoptosis percentage of cultures enriched for RS cells were 2.75-fold and 1.38-fold higher than mMSCs respectively.Conclusion: Glucocorticoid can enhance stem cell self-protection capability,and RS cells have greater capacity to self-protective and self-regulate.
