摘　　要：目的对大鼠NR2B基因核心启动子区进行单核苷酸多态性(single nucleotide polymorphism,SNP)筛查及分析。方法提取16只健康SD大鼠海马组织基因组DNA,聚合酶链式反应(PCR)扩增NR2B基因核心启动子区序列和SNP库已报道的rs8169392所在区域序列,结合DNA测序方法进行SNP筛查和验证。结果在大鼠NR2B核心启动子区705bp中,发现1个新的SNP位点,为-217位C/T多态;本研究中未筛查到NCBI-SNP库报道的rs8169392。结论 -217位C/T多态的发现为NR2B基因多态性数据库提供了新信息。Objective To detect and locate the single nucleotide polymorpbism （SNP） in the core promoter region of NR2B gene of rats. Methods Genomic DNA from hippocami of 16 healthy SD rats was extracted. Polymerase chain reaction was used to clone the core promoter region sequence and rs8169392 region reported in NCBI - SNP database of NR2B gene and DNA sequencing screen SNPs. Results There was 1 SNP in the core promoter region sequence of rat NR2B gene： - 217 C/T. However, rs8169392 polymorphism was not found in this study. Conculsions The finding provides new data for SNP database in the NR2B gene promoter field.
