摘  要:目的观察18F-FDG对基因敲除(Apolipoprotein E-Deficient,ApoE-/-)小鼠主动脉粥样斑块的核素显像,探讨18F-FDG核显像结果与油红染色之间关系,初步评价18F-FDG在检测粥样斑块中的作用。方法 20周龄ApoE-/-小鼠作为实验组,给予高脂饮食(0.15%胆固醇,15%脂肪),于第44周龄前禁食10-12h后,经尾静脉注射(0.56-0.71)mCi 18F-FDG,20min后处死小鼠分离主动脉,将血管纵行剖开,计算主动脉18F-FDG标准摄取值(SUV),然后行放射性自显影、油红染色,比较结果。正常C57BL/6N小鼠为对照组,给予普通饮食,其余做法相同。结果 ApoE-/-小鼠主动脉对18F-FDG的摄取值(0.243±0.054)较对照组C57BL/6N小鼠有显著性差异(0.112±0.004,P<0.05)。放射性自显影示实验组ApoE-/-小鼠主动脉上斑块区域与非斑块区域信号比为5.848(±2.416)∶1,且与油红染色相比其检测斑块的敏感性及特异性分别为60.194%、66.67%;对照组放射自显影图未见有特异性信号浓聚,与之对应的油红染色中亦未见到有染色阳性区域。结论 18F-FDGPET/CT无创性检测动脉粥样斑块具有可行性,有可能成为临床无创评价斑块稳定性的一种方法。Objective To study the application of ^18F-FDG nuclide imaging of atherosclerotic plaques in apolipoprotein E-deficient mouse and the correlation between the ^18F-FDG nuclide imaging and oil red staining.Methods Apolipoprotein E-deficient male mice at the age of 20 weeks were divided into experimental group and control group.Mice in experimental group were fed with Western diet containing 0.15%cholesterol and 15%fat for 44 weeks.After fasted for 10-12 hours,mice were injected with 0.56-0.71 mCi ^18F-FDG into the tail vein,and sacrificed after 20min with their aortas removed and longitudinally incised.The standard uptake value(SUV)of ^18F-FDG was calculated.The radioactivity was measured with a well-typeγ-counter.Aorta plaque and macrophages were investigated with oil red staining.Mice in control group were fed with common diet,and the rest were similar to those in experimental group.Results A significant difference was found in 18F-FDG uptake between the two groups(0.243±0.054 vs 0.112±0.027,P〈0.05).The radioactivity ratio in aortas 20min after 18F-FDG injection was 5.848(±2.416):1.The sensitivity and specificity were 60.194%and 66.67%,respectively.No radioactivity was found in control group.The ^18F-FDG uptake was positively correlated with the number of macrophages(Mf)in thoracic aortic segments.Conclusion 18F-FDG PET/CT can be used in detecting aorta plaques and in evaluating their stability.
