2009年龙岩市人肠道病毒71型VPl基因特征分析

摘　　要：目的分析2009年龙岩市流行的人肠道病毒71(EV71)分离株的分子生物学特征。方法从手足口病例的153份咽拭标本中分离EV71病毒株,选取其中3株,用RT-PCR扩增VP1基因片段,并进行测序和病毒种系进化等分析。结果 3株EV71均为C4基因亚型,与其他省份C4亚型分离株的同源性为94.8%～99.0%;其中EV71/FJ-LY241/09和EV71/FJLY299/09具有更高的同源性,而另1株EV71/FJLY231/09与其他2株距离稍远。氨基酸序列分析表明,3株病毒的VP1具有3个氨基酸位点差异。结论 2009年龙岩市流行的EV71为C4基因亚型,与目前国内其他省份流行的毒株无显著差别,但不同分离株有一定差异,其原因需进一步监测和研究。Objective To analyze molecular biology characteristics of human enteriovirus 71（EV 71） in Longyan city in 2009.Method The virus were isolated form 153 specimens of HFMD（hand-foot-mouth disease）.Three EV 71 isolates were chosen and the VP1 genes were amplified and sequenced by using RT-PCR for sequencing and phylogenetic analysis.Result VP1 genes from three EV 71 isolates were amplified and sequenced.The three EV 71 isolates belonged to C4 subgroup,and the identity with the isolates from other provinces were 94.8%～99.0%.EV 71/FJLY241/09 and EV 71/FJLY299/09 showed higher homologous and the EV 71/FJLY231/09 showed slight heterologous from them.Amino acid sequences analysis showed they had high homologous except the different amino acids exist in three loci.Conclusion The EV 71 epidemic in Longyan in 2009 belongs to C4 sub-genotype.The viruses were basically similar to other provinces,but there are some differences in different isolates and should be studied in the future.
