摘 要:利用PCR-SSCP和测序技术,检测太湖鹅脂肪细胞型脂肪酸结合蛋白(A-FABP)基因内含子2单核苷酸多态性(SNPs)及其对肌内脂肪含量的影响。结果表明:①扩增序列大小为482 bp,内含子2序列大小为174~394 bp,共发现3个突变位点,在187 bp处发生C→T突变,在235 bp处发生C→A突变,在372 bp处发生A→C突变;②AB基因型个体肌内脂肪含量高于AA基因型个体,但差异不显著。该研究结果为进一步分析A-FABP基因的遗传变异及其与肉品质性状的相关性奠定了理论基础。In order to detect the relativity of single nucleotide polymorphism (SNP) of adipocyte fatty acid binding protein (A-FABP) gene with meat quality in the geese, the SNP of A-FABP gene on the sequence sites of the second intro in the Taihu geese was detected with the techniques of PCR-SCP and sequencing. The results showed that: @ The prod uets of PCR were about 482 bp, the intro 2 was from 174 bp to 394 bp and there were three mutational sites, respectively on the 187 bp (C→T), 235 bp (C→A) and 372 bp (A→C) ; (2) The crude fat of AB genotype was higher than that of AA genotype, but the difference was not significant. These findings are good theoretical foundation for the progressive research of the relationship between genetic mutation in A-FABP gene with meat traits.
