摘  要:目的:探讨甲强龙(MP)、电针与羊膜上皮细胞(AECs)移植联合治疗对脊髓损伤(SCI)后大鼠神经纤维再生和功能恢复的作用.方法:成年雌性Wistar大鼠随机分成5组,SCI对照组、甲强龙组、MP+华佗夹脊穴电针组、MP+电针+AECs组和假手术组.各组术后30d神经丝蛋白行(NF)、5-羟色胺(5-HT)和降钙素基因相关肽(CGRP)免疫荧光组织化学观察以及神经电生理检测.结果:MP+电针+AECs组损伤区可见大量有序的NF、5-HT和CGRP阳性神经纤维,其中5-HT阳性纤维假手术组比例高于CGRP阳性纤维所占比例;神经电生理检测显示该组与其他损伤组比较感觉诱发电位与运动诱发电位的峰-峰值增加,潜伏期明显缩短,差异有统计学意义,其中运动诱发电位恢复程度优于感觉诱发电位.结论:甲强龙、电针联合AECs移植治疗脊髓损伤促进了神经纤维的再生和大鼠后肢功能的恢复.Objective: To explore the combined effect of Methylprednisolone (MP), electro-acupuncture (EA) and amniotic epithelial cell (AECs) transplantation on the regeneration of nerve fibers and function recovery for rat with spinal cord injury (SCI). Methods: Sixty female Wistar rats were randomly assigned to 5 groups ( n=12 per group). SCI control group: Improved Allen's bump equipment was employed in the present experiment, and no treatment after SCI was created; MP group: MP pulse treatment after SCI; MP and Huatuo spine points electro-acupuncture group: EA treatment on Hua Tuo Jiaji acupoint after SCI, based on MP group; MP, EA and AECs group: on the basis of MP and Hua Tuo Jiaji acupoint electro-acupuncture, AECs were transplanted with microinjector 7 d after SCI; and Sham group: neural scute was opened without SCI. NF, 5-HT and CGRP immunofluoreseence histochemistry observation and neuro-electrophysiological tests were performed on the 30th day after operation. Results: As revealed by NF, 5-HT and CGRP staining, MP, EA and AECs group contained numerous NF, 5-HT and CGRP-positive nerve fibers. The proportion of 5-HT-positive nerve fibers was higher than CGRP's. Neuroelectrophysiological tests on both somatosensory evoked potential and motor evoked potential demonstrated that peak-to-peak value increased sharply and latency was shortened obviously in MP, EA and AECs group. Difference between MP, EA and AECs groups and other SCI groups was statistically significant, and the recovery of MEP was better than SEP's. Conclusion: A combination of MP, EA and AECs transplantation can promote regeneration of nerve fibers and recovery of hind limb function for SCI rats.
