益生注射液抗大鼠移植心慢性失功加快模型的实验研究  被引量:2

Experimental study on Yisheng injection for a novel accelerated rat cardiac graft failure model induced by ischemia/reperfusion injury

摘  要:目的:探讨中药益生注射液对大鼠移植心慢性失功(Chronic graft failure,CGF)加快模型的保护作用,为临床应用提供部分实验依据。方法:采用雌性封闭群SD大鼠40只,随机分2组,对照组的离体心脏用4℃生理盐水保存3 h后,行心脏移植,术后不给任何药物处理,实验组的离体心脏则用含益生注射液的4℃生理盐水保存3 h后,行心脏移植,术后每天腹腔注射益生注射液,剂量为8 mg/(kg.d),连续9 d。分别采集受体术前和术后1、3、5、7、9 d血液,检测丙二醛(Malondialdehyde,MDA)和超氧化物歧化酶(Superoxide dismutase,SOD)含量变化。另外,在移植术后12周,取2组移植心脏的组织学标本进行比较分析。结果:对照组和实验组心脏移植术后血浆MDA含量显著升高,而SOD活性却显著降低,术后第1天下降最明显。对照组与实验组相比,术前2组血浆的MDA含量和SOD活性相似;术后第1天,对照组血浆MDA含量升高,并明显高于实验组45.27±6.48与32.78±6.61,P<0.05,而SOD活性下降,并明显低于实验组529.77±46.31与663.33±45.92,P<0.05;此后,2组的血浆MDA含量和SOD活性逐渐恢复到正常,但对照组恢复较慢,术后第9天才接近正常,实验组恢复较快,术后第5天已恢复正常。对2组移植心脏的组织学标本进行比较,移植后12周,实验组与对照组相比,移植心慢性排斥程度明显减轻(2.20±0.40与3.6±0.48,F=100.41,P<0.000 1),心肌间质纤维化程度也减轻(1.00±0.32与1.60±0.48,F=21.63,P<0.000 1)。结论:在缺血再灌注损伤(Ischemia reperfusion injury,I/R)诱导的CGF加快模型中,益生注射液对移植心的I/R损伤有保护作用,并能减轻其慢性失功的进程。Objective: To investigate whether the traditional chinese medicine Yisheng injection can reduce the progress of a novel accelerated rat cardiac graft failure model induced by ischemia/reperfusion injury.Methods: 40 female closed colony SD rats were divided into two groups at random.In the control group,the isolated rat hearts were preserved in 0.9% sodium chlorine solution at 4℃ for 3 hours before transplantation,while in the experimental group,the hearts were preserved in 0.9% sodium chlorine solution containing Yisheng injection 0.5 mg/ml at 4℃ for 3 hours before transplantation.After transplantation,the experimental group recipients were treated with Yisheng injection 8 mg/(kg·d) for 9 days.The specimens of blood were harvested from recipients before transplantation and on 1,3,5,7,9 day after transplantation.The changes of MDA and SOD were observed in two groups.Grafts were excised at 12 weeks after transplantation to assess the graft vasculopathy and interstitial fibrosis using HE and Masson staining for two groups.Results:The MDA and SOD of two groups were similar before transplantation.However,the MDA of the two groups increased and the SOD of two groups decreased,especially the first day after transplantation,but it was more significant in the control group.Then,The MDA and SOD of the two groups recovered to normal gradually,but the experimental group recovered to normal on the 5 day after transplantation,faster than the control group,which recovered to normal on the 9 day after transplantation.By 12 weeks,compare the changes of histology of the grafts of the two groups,grafts vasculopathy and interstitial fibrosis were decreased significantly in experimental group.Conclusion: It demonstrated that Yisheng injection can decrease ischemia/reperfusion injury and reduce the development of rat cardiac chronic graft failure in a novel accelerated rat cardiac graft failure model induced by ischemia/reperfusion injury.
