摘 要:为了研究不同地区传染性法氏囊炎病毒(IBDV)毒株的流行特点及遗传变异规律,应用RT-PCR方法对11株IBDV分离株的VP2全基因进行了扩增、克隆,测序后与参考毒株进行核苷酸及其推导氨基酸序列的同源性比对,并绘制遗传进化树。结果表明,其中的9个分离株与国内外IBDV超强毒的核苷酸与氨基酸序列的同源性比较高,并且与超强毒株VP2高变区内的特征性氨基酸相符合,据此可推测近三年流行的毒株以超强毒株为主;分离株CK/SD/GM0911第299位的超强毒株特征性氨基酸由S变为N;分离株CK/SD/LY0810与中等毒力疫苗株2512同属于进化树中的一个分支,并具有相同的特征性氨基酸,两者的核苷酸序列同源性为99.5%,推测CK/SD/LY0810株为中等强毒力毒株。In order to study the characteristics of prevalent strains in different regions and the hereditability and variability of infectious bursal disease virus(IBDV),the full VP2 genes were amplified from the 11 IBDV isolates by RT-PCR respectively,cloned and sequenced.Homologous comparison was carried out and the phylogenetic trees were constructed based on the nucleotide sequences and deduced amino acid sequences from VP2 genes of the reference strains.The results showed that the similarity of the nucleotide sequences and amino acid sequences of VP2 genes was very high compared with that of vvIBDV in China and abroad in 9 of the 11 isolates,and the characteristic amino acids of VP2 hypervariable regions were the same as vvIBDV,so we speculate that the epidemic isolates were almost vvIBDV in the past three years.The site 299 of CK/SD/GM isolate which is a characteristic amino acid of vvIBDV changed from S to N.CK/SD/LY0810 isolate shared a branch and had the same characteristic amino acids with middle virulent vaccine strain 2512,and the similarity of the nucleotide sequences reached up to 99.5%.So we speculate that CK/SD/LY0810 isolate may be a middle virulent strain.
