摘 要:为了获得优良的猪用益生菌株,从断奶仔猪消化道分离一株乳酸菌SIL1,利用16S rRNA技术进行鉴定,研究了菌株的抗逆性,并采用PCR-DGGE及16S rRNA技术分析其对肠道菌群失调小鼠粪便菌群的影响。结果表明,从断奶仔猪回肠内容物分离的菌株SIL1为唾液乳杆菌(Lactobacillus salivarius),耐受pH3和胆盐含量为6 g/L的培养液,能在模拟胃肠液中生长;60℃处理30 min,不影响其生长;对体外共培养的大肠杆菌和沙门氏菌抑菌效果为100%。DGGE图谱显示,肠道菌群失调小鼠经SIL1菌悬液灌胃5~10 d,粪便中的优势条带有所增加;灌胃15 d,优势条带逐渐减少,经序列分析,优势条带与菌株SIL1同源性为99%;同时,菌株SIL1治疗组小鼠体重平均增长率显著高于自然恢复组(P〈0.05)。说明猪源唾液乳杆菌株SIL1具有较强的抗逆性,可定植于小鼠肠道,能有效调节肠道菌群数量的变化并促进小鼠生长,可作为优良的猪用益生菌株。To obtain superordinary probiotic specially for swine,Strain SIL1 was isolated from ileum contents of weaning piglets,identified by physiological and biochemical tests with the analysis of 16S rRNA.The antireversion force of SIL1 were studied.Changes of bacterial flora from feces of alteration of intestinal flora mice were studied after oral administration of SIL1,using techniques of PCR-DGGE and 16S rRNA.The results showed that Strain SIL1 was Lactobacillus salivarius,which could tolerate 6 g/L bile salt and pH 3,and grew in simulated gastrointestinal fluid.Treated with 60℃ for 30 minutes,it made no effect on growth and it had 100% inhibiting rates in vitro antagonism of Escherichia coli O157 and Salmonella.The DGGE profiles from total bacteria of feces showed some new predominant bands appeared after oral administration of SIL1 for 5-10 days.On day 15 after oral administration of SIL1,this new predominant bands almost disappeared except one.Sequence analysis indicated that the new predominant band was Lactobacillus salivarius.The average growth rate of mice treated with SIL1 was significantly higher than spontaneous recovery group(P0.05).This suggests that Lactobacillus salivarius SIL1 from swine has a strong resistance and proliferation in the gastrointestinal tract,and can regulate the intestinal germ community and promote the growth of mice,which can be used as a good probiotic for swine.
