摘  要:目的探讨大黄酸对肥胖糖尿病大鼠脂肪组织抵抗素基因表达及血浆FFA的影响。进而验证大黄酸具有改善胰岛素抵抗的作用。方法小剂量链脲佐菌素加高脂饲料诱导建立肥胖糖尿病大鼠模型,成模大鼠随机分为模型组、吡格列酮组和大黄酸组。正常组与模型组分别予生理盐水10 ml·kg^(-1)·d^(-1),吡格列酮组予吡格列酮10 mg·kg^(-1)·d^(-1),大黄酸组予大黄酸100 mg·kg^(-1)·d^(-1),分别灌胃8周。测定各组附睾脂肪垫抵抗素基因的表达、血浆FFA及血脂水平。结果与模型组相比,大黄酸组与吡格列酮组脂肪组织抵抗素的表达均下降,FFA、TC、TG、LDL-C均明显降低(P均<0.05)。结论大黄酸能降低肥胖糖尿病大鼠脂肪组织抵抗素表达,降低血浆FFA。这可能是大黄酸改善胰岛素抵抗的机制之一。Objective To investigate the effects of rhein on the expression of resistin mRNA and plasma free fatty acid(FFA) and the association between the effects and insulin resistance in diabetic obese rats. Methods The streptozotocin (STZ)- and high fat diet-induced diabetic obese rats were divided into three groups: diabetic control (DM), pioglitazone (Pio) and rhein (Rh) treatment groups in addition to normal rats. The expression of resistin mRNA, plasma free fatty acid (FFA), and blood lipid were detected after drug or normal saline administration by gavage for 8 weeks. Results The groups of rhein and pioglitazone treatment reduced the expression of resistin mRNA and the level of FFA, TC, TG and LDL as compared with untreated diabetic obese rats (P〈0. 05). Conclusion Rhein therapy can reduce the resistin retiNA expression of adipose tissue and decrease plasma free fatty acid (FFA), which may be one of mechanisms by which rhein can improve IR.
