摘 要:以亚心形扁藻(Platymonas subcordiformis)、大溪地球等鞭金藻(Isochrysis galbana daxidi)和小新月菱形藻(Nitzschia closterium)为实验对象,探讨批次培养过程中不同生长阶段及不同温度处理的藻细胞总ATP酶活性。结果表明:三种微藻在不同生长阶段的细胞总ATP酶活性均有显著变化。同一种微藻生长繁殖越快的阶段,其相对生长常数K和藻细胞总ATP酶活性也越大。不同生长阶段亚心形扁藻、大溪地球等鞭金藻和小新月菱形藻的相对生长常数K与其细胞总ATP酶活力均具有显著的正相关(P〈0.05),其相关系数r分别为0.92、0.79、0.82。亚心形扁藻在25~27℃之间细胞总ATP酶活性无显著变化,在30℃以上细胞总ATP酶活性显著下降。大溪地球等鞭金藻在25~30℃,细胞总ATP酶活性随温度的升高而显著升高,温度高于30℃细胞总ATP酶活性则急剧下降。小新月菱形藻在经20℃以上温度处理后,细胞总ATP酶活性总体上随温度的升高而显著降低。本研究表明藻细胞的总ATP酶活性与微藻细胞生长活力密切相关,并可应用于微藻适宜培养温度的筛选。The paper dealed with the total ATPase activity of Platymonas subcordiformis,Isochrysidaceae and Nitzschia closterium at different growth phases in batch culture and assayed the total ATPase activity of the microalgae experienced different heat treatments.Results indicated that the total ATPase activity of the microalgae cell at different growth phases were significantly different.Higher ATPase activity and higher relative growth constants(K) were detected in the faster growth phase of the same microalgae.There were significant postive correlations between the relative total ATPase activity and the growth constants of microalgae at the same growth phase.The correlation coefficients for Platymonas subcordiformis,Isochrysidaceae and Nitzschia closterium were 0.92,0.79 and 0.82 respectively.There were no significant changes in the total ATPase activity of Platymonas subcordiformis at temperatures ranging from 25 ℃ to 27 ℃.While the total ATPase activity decreased significantly when Platymonas subcordiformis were treated with temperature more than 30 ℃.The total ATPase activity of Isochrysis galbana daxidi increased greatly with an increasing temperature between 25 ℃ and 30 ℃,but declined sharply when heat-treatment temperature were higher than 30 ℃.The significant lower total ATPase activity of Nitzschia closterium were detected at higher temperature(20 ℃).It is therefore suggested that the ATPase activity of the microalge cell was correlated with its viability closely,and be applied to screen suitable culture temperature of microalgae.
