摘  要:目的观察缺血后处理(IPo)对大鼠后肢缺血/再灌注(I/R)后肝脏的保护作用。方法建立SD大鼠后肢I/R模型,将32只健康SD雄性大鼠随机分为4组,每组8只。检测各组肝功能;肝组织中超氧化物歧化酶(SOD)、丙二醛、髓过氧化物酶(MPO)的含量变化;肝组织中Bcl-2和Bax蛋白的表达水平;肝脏细胞凋亡情况。结果与I/R组相比,IPo组及缺血预处理(IPC)组血清肝酶的水平明显降低(P<0.O5);肝组织丙二醛、MPO含量显著降低(P<0.05),而SOD含量明显升高(P<0.05);Bcl-2表达升高,Bax表达降低,凋亡指数下降,差异均有统计学意义(P<0.05)。而IPo组与IPC组相比无显著性差异(P>0.05)。结论 IPo对下肢I/R损伤后引起的肝脏损伤具有明显的保护作用。Objective To observe the effect of ischemic postconditioning on the injury of liver function after ischemia/reperfusion(I/R) in rat hind legs.Methods SD rat hind leg I/R model is set up with 32 healthy male SD rats randomly divided into 4 groups of 8.The hepatic function and the content changes of SOD,malondialdehyde and MPO in hepatic tissue were observed;the expressions of Bcl-2 and Bax in hepatic tissue were measured;the apoptosis in hepatic tissue was assessed.Results Compared with I/R,the activity of plasma enzyme and the concentrations of malondialdehyde and MPO in hepatic tissue in IPC and IPo decreased sharply(P〈0.05),while the concentrations of SOD increased significantly(P〈0.05).Moreover,the expression of Bcl-2 increased;the expression of Bax and the apoptotic index decreased,all of which are statistically different(P〈0.05).The difference between IPo and IPC was not obvious(P〈0.05).Conclusion IPo has obvious protective effects on the liver function after ischemia/reperfusion in rat hind legs.
