摘 要:目的:考察甲磺酸帕珠沙星搽剂(PL)对痤疮丙酸杆菌的体外抑制情况以及对兔耳微痤疮模型的影响。方法:微量稀释法测定PL对痤疮丙酸杆菌标准菌株及临床分离菌株的最小抑菌浓度(MIC);另用涂抹油酸及皮下注射痤疮丙酸杆菌建立微痤疮模型兔耳,造模后第14天随机分为模型对照组、溶媒对照组、阳性对照组(痤康王0.1mL·kg-1)和PL高、低剂量组(0.3、0.1mL·kg-1),每组6只,右外耳道涂擦相应药物,连续14d,末次给药后18h处死,考察各组兔耳组织学变化及病理评分,并设立空白对照组。结果:PL对受试菌株的MIC均为1.0mg·L-1;与模型对照组比较,阳性对照组和PL高、低剂量组兔耳肉眼可见局部毛囊角栓减少,丘疹变平、减少,表皮增厚脱落不明显;组织学观察可见真皮炎症细胞明显减少,毛囊口扩张减轻;病理评分明显降低(P<0.01)。结论:PL对痤疮丙酸杆菌有明显体外抑制作用,同时对微痤疮模型兔耳有治疗作用。OBJECTIVE:To investigate the inhibition effect of Pazufloxacin mesilate liniment(PL) on Propionibacterium acnes(P. acnes) in vitro and rabbit ear acne model. METHODS:MIC of PL against P. acnes standard strains and clinical isolated strains were determined by microdilution method. Rabbit ear acne model was established by spreading oleic acid and injecting with P. acnes subcutaneously. 14 days after modeling,the rabbits were randomly divided into model control group,solvent control group,positive control group(0.1 mL·kg-1 clindamycin and metronidazole liniment) and high-dose and low-dose PL groups(0.3 mL·kg-1,0.1 mL·kg-1) with each group of 6 rabbits. Relevant drug was spreading on the right meatus of external ear for 14 days. Those rabbits were sacrificed 18 hours after the last administration. Histology change and pathologic score were investigated and blank control group was set up. RESULTS:MIC was 1.0 mg·L-1. Compared with model control group,papules and local folliclular plugging in positive control group and high-dose and low-dose groups were all decreased while the incrassation and exfoliation of epidermis was not obvious. The inflammatory cells of dermis were also decreased,and the dilatation of pilosebaceous orifice relieved. Pathologic score decreased significantly(P0.01) . CONCLUSION:PL can inhibit the growth of P. acnes significantly in vitro,and has therapeutic effect on rabbit ear acne.
