摘  要:目的:采用核磁共振(MRI)动态观测白藜芦醇预处理对大鼠局部脑缺血4h和24h时脑水肿和梗死脑组织体积的影响。方法:20只SD大鼠随机分为白藜芦醇预处理组(简称预处理组,n=10)和对照组(n=10)。预处理组手术前10天,每日按照30 mg/kg腹腔注射白藜芦醇溶液。对照组给予同等体积的白藜芦醇溶剂注射10天。10日后行左侧大脑中动脉永久缺血术(MCAO)。术后4 h和24 h分别采用MRI对大鼠纹状体区和顶颞叶区进行T1-SE、T2-TSE、T2-map-SE以及DW-EPI序列的扫描。采用ImageJ软件分析扫描图像中4个感兴趣区域(左侧纹状体、顶颞叶,右侧纹状体、顶颞叶)的表观弥散系数(ADC)值、T2横向弛豫时间(T2-relaxation time,T2RT)值的变化。术后24 h处死大鼠,TTC(2,3,5-氯化三苯基四氮唑)染色脑片,UTHSCSA Image Tool软件计算梗死脑组织体积的比例。结果:①预处理组大鼠缺血4 h时纹状体和顶颞叶相对ADC值较对照组显著增加(P<0.01),左侧T2RT值显著减小(P<0.05)。②预处理组大鼠缺血24 h时纹状体和顶颞叶相对ADC值较对照组均显著增加(P<0.01),左侧T2RT值也显著减小(P<0.05)。③预处理组大鼠梗死组织脑体积比例为0.313±0.047,对照组为0.396±0.021。两组相比有显著性差异(P<0.01)。结论:白藜芦醇预处理对大鼠脑缺血后4 h和24 h时均有减轻脑水肿的作用,并且可以有效减少脑梗死体积,从而起到神经保护作用。Objective To dynamically observe the effects of resveratrol preconditioning on the focal cerebral ischemia.Methods Twenty SD rats were randomly divided into the resveratrol preconditioning group(n = 10)and the control group(n = 10).The rats in the preconditioning group received intraperitoneal injection of resveratrol solution with the dosage of 30mg/kg per day for 10 days before the surgery.The rats in the control group received the same dose of resveratrol solvent injection for 10 days.Then,the left middle cerebral artery of all rats was occluded permanently(MCAO).At 4 hours and 24 hours after MCAO,the striatum and parietotemporal cortex were scanned by MRI with the sequence of T1-SE,T2-TSE,T2-map-SE and DW-EPI.Four regions of interest(ROI),including ipsilateral striatum,ipsilateral parietotemporal cortex,contralateral striatum and contralateral parietotemporal cortex were analyzed by ImageJ software on the scanned images and thus the apparent diffusion coefficient(ADC) value and T2 transverse relaxation time(T2RT) value were obtained.All rats were sacrificed at 24 hours after MCAO.UTHSCSA ImageTool software was used to calculate the infarct volume ratio on the brain slices stained with TTC(2,3,5-triphenyl tetrazolium chloride).Results ①The relative ADC value of the striatum and parietotemporal cortex significantly increased(P 0.01) and T2RT significantly decreased(P 0.05)of the rats in preconditioning group as compared to the control group at 4 hours after the MCAO.②Similarly,the relative ADC value of the striatum and parietotemporal cortex also significantly increased(P 0.01) and the T2RT significantly decreased(P 0.05) of the rats in preconditioning group as compared to the control group at 24 hours after MCAO.③ The infarction volume ratio of preconditioning group and control group was 0.313±0.047 and 0.396±0.021,respectively.There was a significant difference between the two groups.Conclusion Resveratrol preconditioning reduced the cerebral edema and infar
