补肾活血针刺法对阿尔茨海默病模型小鼠海马脑红蛋白表达的影响

摘　　要：目的探讨补肾活血针刺法对阿尔茨海默病模型快速老化小鼠P8(SAMP8)学习记忆能力的影响及作用机制。方法将20只5月龄雄性SAMP8小鼠随机分为针刺治疗组、模型对照组各10只,另设5月龄雄性抗快速老化小鼠(SAMR1)10只作为正常对照组。针刺治疗组予以补肾活血针刺法干预,选穴百会、血海、肾俞、膈俞,采用捻转补泻手法。模型对照组及正常对照组未予特殊处理。干预4周后,取各组小鼠海马组织,采用免疫组化法检测脑红蛋白(Ngb)的表达情况。结果模型对照组小鼠海马组织Ngb的表达水平较正常对照组减弱(P<0.01);与模型对照组相比,针刺治疗组小鼠海马组织Ngb的表达水平明显升高(P<0.01)。结论补肾活血针刺干预能够明显增强SAMP8小鼠海马组织Ngb的表达,这可能是其改善SAMP8小鼠学习记忆能力的潜在分子机制之一。Objective To investigate the effect and mechanism of acupuncture for reinforcing kidney and activating blood on learning and remembering ability of senescence-accelerated mouse P8（SAMP8） models of Alzheimer＇s disease.Methods Twenty 5-month-old male SAMP8 mice were randomized into two groups,acupuncture treatment group and model control group,10 in each.Another ten 5-month-old male senescence-accelerated mouse/resistance 1（SAMR1） were prepared as normal control group.The acupuncture treatment group was treated by acupuncture with reinforcing kidney and activating blood method,the selected points were Baihui（GV20）,Xuehai（SP10）,Shenshu（BL23）,and Geshu（BL17）,and rotating the needles for reinforcing-reducing was adopted in the manipulation.The model control group and normal control group were not given special treatment.After 4 weeks,the neuroglobin（Ngb） expression in the hippocampus tissues of mice of all groups was examined by immunohistochemical method.Results The Ngb expression of the model control group was weaker than that of the normal control group（P0.01）.Compared with the model control group,the Ngb expression of the acupuncture treatment group was significantly increased（P0.01）.Conclusion Acupuncture invention can strengthen the Ngb expression in hippocampus tissue of SAMP8 mice.It＇s possibly one of the underlying molecular mechanisms of acupuncture in improving learning and memory abilities of SAMP8 mice.
