摘  要:目的:探讨山楂叶总黄酮对大鼠脑缺血再灌注损伤的保护作用机制。方法:采用改良Zea Longa’s法制作大脑中动脉闭塞(MCAO)模型。将健康雄性SD大鼠随机分成假手术组、模型组、尼莫地平组、山楂叶总黄酮(HLF)低、中、高剂量组6组。用神经功能评分评价大鼠脑缺血再灌注后行为障碍;用2,3,5-三苯基氯化四氮唑(TTC)染色法测定梗死灶面积;用相应试剂盒测定脑组织内白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)。结果:模型组缺血脑组织内的IL-1β、TNF-α含量显著升高,与假手术组相比差异有显著性,尼莫地平组和HLF组能明显降低缺血脑组织内IL-1β、TNF-α含量,与模型组相比有显著差异,HLF高剂量组和尼莫地平组缺血脑组织内IL-1β、TNF-α含量与HLF与低剂量组相比也有显著差异。结论:HLF可能通过抑制缺血再灌注后炎症反应过程而起到抗脑缺血再灌注损伤作用。OBJECTIVE To investigate the effects of hawthorn leaves flavonoids (HLF) on cerebral ischemia-reperfusion injury in rats, and to elucidate the neuroprotective machanisms of HLF. METHODS The model of focal ischemic reperfusion in Sprague-Dawley (SD) rats was prepared by middle cerebral artery occlusion (MCAO) with a nylon monofilament suture. Adult male SD rats were divided randomly into sham operation group, model group, nimodipine treatment group, HLF treatment group (low dosage, middle dosage, high dosage). The behavioral disturbance was evaluated according to neurological deficit scores, the infarction volumes were evaluated by 2,3,5-triphenyltetrzolium chloride (TTC) staining, the levels of MDA, SOD, NO in the brain tissues of ischemia-reperfusion rats were measured by the corresponding kits. RESULTS Compared with the sham operation group, model group's IL-1β, TNF-α increased significantly. Compared with the model group, nimodipine and HLF groups' IL-1β, TNF-α decreased significantly. As the dosage of HLF increasing, IL-1β, TNF-α decreased significantly. Compared with HLF-L group, HLF-H and nimodipine groups' IL-1β, TNF-α decreased significantly. CONCLUSION HLF has inhibitory effects on inflammation caused by ischemia-reperfusion, and then protects cerebral ischemic injury.
