摘 要:目的观察硫酸铟对中国仓鼠肺成纤维细胞(V79)的DNA损伤作用。方法以体外培养的V79细胞为研究对象,四氮唑蓝比色分析法(MTT法)观察不同染毒浓度(0.5、1.0、2.0、4.0、8.0 mmol/L硫酸铟)和不同染毒时间(2、6、12、24 h)的细胞毒性;应用彗星实验(单细胞凝胶电泳技术)检测经不同浓度的硫酸铟染毒不同时间后对V79细胞DNA的损伤作用。结果当硫酸铟浓度≥0.5 mmol/L时,各染毒组细胞存活率与对照组比较降低,差异有统计学意义(P<0.05)。各染毒组拖尾率与阴性对照组比较,差异有统计学意义(P<0.05),且随着硫酸铟浓度的增加,呈上升趋势;Tail DNA%,尾长,Olive尾矩随染毒剂量增大而增加,与阴性对照组比较,差异有统计学意义(P<0.05)。结论硫酸铟可以引起V79细胞急性毒性,细胞存活率呈浓度-时间依赖性下降。硫酸铟对V79细胞的DNA有损伤作用,且有明显的剂量-效应关系。Objective To investigate the cytotoxicity and mechanism of indium sulfate on V79 cells in vitro.Methods V79 cells cultured in vitro were taken as the subject which were exposed to indium sulfate at different dosages(0.5,1.0,2.0,4.0 and 8.0 mmol/L)for different exposure periods(2,6,12 and 24 h).The cytotoxicity was observed with MTT colorimetric assay.SCGE was used to detect the DNA damage of V79 cells after exposed to indium sulfate of different concentrations for different hours.Results When the concentration of indium sulfate was≥0.5 mmol/L,the survival rates of V79 cells were inhibited in all exposure groups,(P0.05).The cell decreased with the increase of the dosage and time showing a dose-dependent and time-dependent relationship.The damaged cell rate,tail length,Olive tail moment and tail DNA% of five exposure groups increased significantly with the increase of indium sulfate concentration.And the statistic differences between the negative groups were significant(P0.05).All the analytical indexes including tail length,Olive tail moment and tail DNA% increased in a dose-dependent pattern.Conclusions Indium sulfate could affect the V79 cell viability and obviously inhibit its growth.It might correlate with the DNA damage.
