摘 要:【目的】探明甘蔗黄叶病在广西北部蔗区的发生情况,为甘蔗健康种苗生产提供科学依据。【方法】从广西北部蔗区采集不同品种显症或不显症甘蔗样品,采用引物(P1:5′-AATCAGTGCACACATCCGAG-3′/P2:5′-GGAGCGTCGCCTACCTATT-3′)进行SCYLV一步法RT-PCR检测。【结果】柳州郊区蔗区调查的8个品种中有6个品种带毒,带毒率为75.0%;柳城蔗区调查的7个品种全部带毒,带毒率为100.0%;鹿寨蔗区调查的4个品种中有2个品种带毒,带毒率为50.0%;宜州蔗区调查的6个品种中有4个带毒,带毒率为66.7%。在所调查的品种中,柳城03-182、桂糖21和台糖16在作为调查对象的蔗区均检测出黄叶病毒;主栽品种ROC22也在柳城和宜州蔗区检测出黄叶病毒。【结论】甘蔗黄叶病已在广西北部蔗区发生,且有向外蔓延的趋势。[Objective]The present study was conducted to find out the occurrence of sugarcane yellow leaf virus(SCYLV) disease in the north sugarcane growing areas of Guangxi, and to provide scientific basis for healthy seedcane production of sugarcane. [Method]Sugarcane leaf samples with different stages of symptoms were collected from Liuzhou suburbs (8 samples), Liucheng (7 samples), Luzhai (4 samples) and Yizhou (6 samples) areas of Guangxi. The virus was detected using one-step RT-PCR method with SCYLV specific primers (P1:5′-AATCAGTGCACACATCCGAG-3′/P2:5′-GGAGCGTCGCCTACCTATT-3′). The amplified cDNA products were sequenced. [Result]Six out of 8 varieties (occupying about 75%) were found infected with SCYLV in Liuzhou suburbs and the rate of SCYLV infection in other three towns, viz., Liucheng, Luzhai and Yizhou were 100.0, 50.0 and 66.7%, respectively. In addition, the varieties, GT21, Liucheng 03-182 and ROC16 were found infected by SCYLV in the tested areas. The major cultivated sugarcane variety ROC22 also gave positive results of SCYLV test in Liuzhou and Yizhou areas. [Conclusion]Sugarcane yellow leaf disease was found to occur in northern areas of Guangxi, efforts must be done to develop proper measures to control the spread of this disease.
