去整合素Kistrin对晶状体摘除术后兔晶状体上皮细胞MMP-2和TIMP-2基因表达的影响

摘　　要：目的研究去整合素Kistrin作用下,晶状体摘取术后兔晶状体上皮细胞(LECs)中基质金属蛋白酶2(MMP-2)mRNA和其抑制因子组织性金属蛋白酶抑制物2(TIMP-2)mRNA表达的变化,探讨Kistrin作用的分子机制。方法将8只新西兰大白兔随机分为实验组和对照组,每组4只,均行右眼透明晶状体囊外摘除术(ECLE)建立后嚢膜混浊腜?PCO)。术毕实验组囊袋内注入浓度为80μg/L的Kistrin 0.2 ml,对照组注入等量林格液,随机选4只左眼为空白组。术后1 d、2 d、3 d、7 d、14 d在裂隙灯下观察实验动物眼部情况。术后14 d后取嚢膜行实时荧光定量PCR检测MMP-2 mRNA和TIMP-2 mRNA的表达量。结果空白组后嚢膜上MMP-2 mRNA和TIMP-2 mRNA表达分别为(0.127±0.039)、(2.485±0.507)。术后14 d对照组两个基因表达量分别为(3.785±0.934)、(19.903±10.499),均高于空白组(P<0.05)。实验组MMP-2 mRNA的表达量为(0.401±0.253),低于对照组(P<0.05),而与空白组比较差异无统计学意义(P>0.05)。实验组TIMP-2mRNA的表达量为(16.119±3.773),与对照组比较差异无统计学意义(P>0.05),但高于空白组(P<0.05)。结论晶状体囊外摘除术后使用Kistrin,MMP-2 mRNA表达在正常水平,TIMP-2 mRNA的表达则仍然维持术后高水平,可能是减少IV型胶原分泌和LECs增殖的关键因素。Objective To investigate the regulation of MMP-2 mRNA and TIMP-2 mRNA expression by disintergrin Kistrin in lens epithelial cells（LECs）and explore the mechanisms of Kistrin.Methods 8 New Zealand rabbits were randomly divided into control group and experimental group,4 cases each group.Extracapsular lens extraction was performed on right eye of each rabbit to establish the model of posterior capsular opacification.0.2 ml 80 μg/L Kistrin was injected into the capsular bag of lens in experimental group and the normal saline 0.2 ml in the control group just after the operation.4 left eyes were chosen randomly as blank control group.All eyes were observed by slit-lamp microscopy at the 1st day,2nd day,3rd day,7th day and 14th day.14 days after the operation,capsule membranes were taken out and the expression of MMP-2 mRNA and TIMP-2 mRNA were detected by real-time RT-PCR.Results MMP-2 mRNA and TIMP-2 mRNA could be detected in normal rabbit LECs,and the abundance were（0.127±0.039）,（2.485±0.507）,respectively.The abundance of MMP-2 mRNA was（3.785±0.934）and that of TIMP-2 mRNA was（19.903±10.499）on 14th day after surgery.compared with the blank control group,the expression of MMP-2 mRNA and TIMP-2 mRNA increased significantly（P0.05）.In the experimental group,the abundance of MMP-2 mRNA was（0.401±0.253）,which decreased obviously compared with the control group（P0.05）.There were no significant difference between experimental and blank group（P0.05）.The abundance of TIMP-2 mRNA was（16.119±3.773）inexperimental group and there were no significant difference between experimental and control group in the expression of TIMP-2 mRNA（P0.05）,but compared with the blank control group,it increased obviously（P0.05）.Conclusion Disintergrin kistrin could keep the expression of MMP-2 mRNA normal and increase the expression of TIMP-2 mRNA after extracapsular lens extraction,which may be a key factor of the decrease of type Ⅳ collagen secretion and the proliferation of LECs.
