利用电转染技术对大鼠原代系膜细胞进行Septin2质粒和siRNA转染的研究

摘　　要：目的:利用电转染技术,对大鼠原代系膜细胞进行Septin2质粒和siRNA转染,制备出Septin2过表达和低表达的细胞模型。方法:培养原代系膜细胞,利用电转染方法,转染pEGFP-C1、pRK5和pRK5-Septin2质粒,以及Septin2的siRNA。结果:通过荧光显微镜观察,pEGFP-C1转染的阳性率可达40%,pRK5-Septin2质粒转染组Septin2表达显著升高,而Septin2siRNA转染组Septin2表达显著下降。结论:利用电转染方法转染Septin2质粒和siRNA,成功的建立了Septin2过表达和低表达的细胞模型,为日后研究Septin2的功能建立基础。Objective：Cell model with Septin2 overexpression or inhibition was set up by electroperation of Septin2 plasmid or siRNA.Methods：Rat primary mesangial cells cultured were transfected with pEGFP-C1、pRK5,pRK5-Septin2 and siSeptin2.Results：The results showed that the positive rate of pEGFP-C1 transfection was 40% when detected by fluorescence microscopy;there was significantly higher expression of Septin2 in SEPT2-transfected cells and lower expression of Septin2 in siSEPT2-transfected cells.Conclusion：We set up cell model with Septin2 overexpression and inhibition successfully by electroperation of Septin2 plasmid or siRNA,which was essential of further research on the function of Septin2 in the mesangial cells.
