摘 要:目的探讨糖尿病胰腺组织miRNA的表达变化。方法链脲佐菌素(STZ)40 mg/kg腹腔注射+高脂高糖饲料喂饲健康雄性SD大鼠,成模(测血糖≥16.7 mmol/L)后4周,取胰腺组织常规抽提总RNA,采用含有2340种miRNA的芯片检测系统进行miRNA表达谱差异分析。结果模型组miRNA表达谱中异常表达(2倍以上)的miRNA共有37个。其中,17个表达上调,20个表达下调。结论在大鼠糖尿病模型中,部分胰腺miRNA的表达发生明显变化,提示miRNA可能参与糖尿病诱发的调节作用。Objective To investigate the expression pattem of microRNA(miRNA) in the pancreas of type 2 diabetes mellitus(T2DM) by miRNA array technique.Methods T2DM rat model(blood glucose16.7 mmol/L) was established in male SD rats by intraperitoneal injection of STZ 40 mg/kg and feeding high sugar and fat for 4 weeks.After the rats were sacrificed,total RNA was extracted from the pancreas and the differential expression of miRNA was detected by miRNA array.Results Compared to the rats in normal control group,abnormal expression of miRNA(the change was greater than two times) was seen in 37 miRNAs in the rats of model group,of which 17 miRNAs were up-regulated and the other 20 miRNAs were down-regulated.Conclusion In the pancreas of T2DM rat model,the expression of miRNA is changed,which implies that those changed miRNAs may play an important role in development of diabetes in SD rats.
