摘  要:目的:探讨Bc l-2、Fas基因在白毛藤总苷诱导人肝癌Bel7402细胞凋亡中的作用。方法:荧光显微镜观察不同浓度白毛藤总苷诱导Bel7402细胞凋亡作用情况,RT-PCR检测Bcl-2、Fas基因表达量的变化。结果:白毛藤总苷诱导Bel7402细胞凋亡,并且上调Fas和降低Bcl-2表达。结论:白毛藤总苷能促进Bel7402细胞凋亡,其机制可能与激活Fas、抑制Bcl-2表达有关。Objective:To investigate the effect of Bcl-2 and Fas gene in Solamum lyratum Thunb total glycosides inducing apoptosis of Human Hepatocellular carcinoma cell line Bel7402 and the molecular mechanism.Methods:Bel7402 cells were treated with various Solamum lyratum Thunb total glycosides,We observed cell apoptosis by fluorescence microscope.We detected the levels of gene expression in all groups by RT-PCR.Results:Solamum lyratum Thunb total glycosides have obviously apoptosis-inducing effects on Bel7402 cells and could effectively decrease the level of Bcl-2 gene expression and improve the level of Fas gene expression.Conclusion:Solamum lyratum Thunb total glycosides could induce apoptosis of Bel7402 cells probably through down regulating Bcl-2 gene expression and up regulating Fas gene expression.
