摘  要:目的:观察首乌延寿丹对兔动脉粥样硬化(AS)形成的防治作用及其可能的机制。方法:将32只日本大耳白兔随机分成空白对照组、模型组、首乌延寿丹组、辛伐他汀组,高脂饲料喂养制作AS模型。第12周末测定血脂变化,观察主动脉病理形态学改变,免疫组化法测定血小板源生长因子-B(PDGF-B)、基质金属蛋白酶-9(MMP-9)蛋白表达。结果:以高脂饲料喂养成功建立AS模型,PDGF-B、MMP-9表达明显升高;首乌延寿丹组及辛伐他汀组AS病变均明显减轻,PDGF-B、MMP-9蛋白表达明显下降(P<0.01),首乌延寿丹组与辛伐他汀组比较无明显差异(P>0.05)。结论:首乌延寿丹可明显抑制AS发展,这一作用可能是通过抑制PDGF-B、MMP-9蛋白表达来实现的。Objectives: To investigate the preventive effects of SWYSD on the rabbits with atherosclerosis(AS).Methods: 32 Japan big-ear white rabbits were randomly divided into normal control group﹑model group﹑SWYSD group﹑Simvastatin group.The AS model was established with high fat diet.Serum lipid level was measured﹑pathological changes of aorta was analyzed and the expression of platelet-derived growth factor-B(PDGF-B)、matrix metalloproteinase-9(MMP-9) was detected by immunohistochemistry in the 12th week.Results:The AS model was success fully established with high fat diet.There was severe pathological lesion in model group,the expression of PDGF-B﹑MMP-9 in model group were upregulated(P0.01).AS was less severe in the SWYSD group and Simvastatin group;the expression of PDGF-B﹑MMP-9 were downregulated.There was no significant difference between SWYSD group and Simvastatin group(P0.05).Conclusion: SWYSD and Simvastatin can inhibit the expression of PDGF-B and MMP-9,this effect maybe one of the key factors in preventing the formation of AS.
