2型糖尿病大鼠不同组织中p85mRNA及蛋白表达变化与中药干预作用

摘　　要：目的:观察2型糖尿病大鼠不同组织中p85基因表达的变化及复方黄连降糖片对其变化的影响。方法:对链脲佐菌素注射加高脂饮食诱导的2型糖尿病模型大鼠给予复方黄连降糖片治疗8周。测定血糖、血清胰岛素,大鼠肝脏、骨骼肌和脂肪组织p85mRNA的表达及其蛋白的表达。结果:复方黄连降糖片治疗组大鼠的血糖和血清胰岛素均有所下降,各组织p85的基因表达均显著提高。结论:不同组织p85的基因表达的提高可能是复方黄连降糖片治疗2型糖尿病的分子机理之一。Objective： to investigate of the Variation of Gene Expression of p85 of Type 2 Diabetes Mellitus Rats in Different Tissue and Intervention of Fufang Huanglian Jiangtangpian（abbr：FFHLJTP） on these indexs.Methods： FFHLJTP was given to type 2 diabetic rats.Blood glucose was measured.Blood insulin was measured.The expression of p85mRNA and protein in liver,muscle and fat of type 2 diabetic rats ware determined by RT-PCR and western blot analysis.Results： FFHLJTP treatment decreased the density of blood glucose and blood insulin in type 2 diabetic rats.The mRNA and protein expressions of p85 in liver,muscle and fat were obviously increased.Conclusion：FFHLJTP increases p85mRNA and protein expression in liver,muscle and fat of type 2 diabetic rats.Its mechanism may be partly by raising p85 gene expression in liver,muscle and fat.
