固本活血壮骨颗粒对去卵巢大鼠血清IGF-1和IL-1β影响的实验研究

摘　　要：目的:通过观察固本活血壮骨颗粒对实验性骨质疏松症大鼠血清胰岛素样生长因子-1(IGF-1)、白介素1-β(IL-1β)水平的影响,探讨固本活血壮骨颗粒防治骨质疏松症的微观机制。方法:取3月龄的雌性SD大鼠60只,随机分为假手术组、模型组、仙灵骨葆胶囊组、尼尔雌醇组、固本活血壮骨颗粒小剂量组、大剂量组6组。除假手术组仅切除双侧卵巢附近的相当于卵巢大小的小块脂肪外,其余各组均切除双侧卵巢。术后1周开始给药,灌胃90d后取血分离血清,采用ELISA法检测血清IGF-1、IL-1β水平。结果:模型组与假手术组比较,IGF-1水平降低,IL-1β水平增高,且差异有显著性(P<0.05);仙灵骨葆胶囊组和大剂量组提高IGF-1水平,降低IL-1β水平,与模型组比较有显著性差异(P<0.05)。结论:固本活血壮骨颗粒通过提高IGF-1水平、降低IL-1β水平,改善去卵巢大鼠骨组织显微形态,对去卵巢大鼠的骨质疏松症具有防治作用,此作用具有一定的剂量依赖性。Objective：To explore the molecular mechanism of traditional medicine GubenHuoxueZhuanggu Granules on rats with Osteoporosis via measuring the level of IGF-1 and IL-1β in serum.Methods：Sixty three months old female SD rats were randomly divided into six groups（n=10）：sham operation group,model group,XianlingGubao Capsule group,Nylestriol group,low-dose group and high-dose group of the traditional medicine GubenHuoxueZhuanggu Granules.A part of fat near the bilateral was removed from the rats of the sham operation group,while the bilateral ovaries of the rats in other groups were removed.After one week,the rats of the sham operation group and the model group were given normal saline（10 ml/kg）,the rats of the XianlingGubao Capsule group were given the capsules called XianlingGubao（0.469 g/kg）,the rats of the Nylestriol group were given Nylestriol（10ml/kg）,while the rats of the low-dose group and the high-dose group of the traditional medicine GubenHuoxueZhuanggu Granules were given the granules of low-dose（0.938g/kg）and high-dose（4.69g/kg）respectively.The method of giving medicine was intragastric administration.After 90 days with intragastric administration,blood was collected from their hearts and serum Was separated.The level of IGF-1 and IL-1β were measured via the method of ELISA.The upper parts of the right shins of rats were made into pathologic slices.The differences of shapes and cellular morphology between of the pathologic slices of different groups were found via HE Staining.Results：Model group compared with sham operation group showed the lower level of IGF-1,IL-1β increased levels,and there wasf a significant difference（P0.05）.Each group compared with model group increased IGF-1,IL-1β reduced.Nylestriol group and low-dose group compared with model group showed non-significance difference（P0.05）.XianlingGubao Capsule group and high-dose group compared with model group have significant difference（P0.05）.Conclusion：The traditional medicine GubenHuoxueZhuang
