摘 要:目的观察不同浓度的氯化两面针碱对斑马鱼心脏的影响。方法用发育正常的受精后48 h(48 hpf)的斑马鱼胚胎作为实验模型,用浓度为5.00 mg/L、3.15 mg/L、2.00 mg/L、1.58 mg/L、1.12 mg/L的氯化两面针碱处理上述胚胎,分别于受精后60 h和72 h时观察胚胎心脏的形态及心率的变化。结果各给药组在受精后60 h和72 h时均出现胚胎心脏中毒现象。5.00 mg/L、3.15 mg/L实验组中,胚胎主要表现为心脏跳动停止、心脏区域出血,2.00 mg/L、1.58 mg/L、1.12 mg/L实验组的胚胎则主要表现为心脏发育畸形。心率随着浓度的升高而明显下降,随着暴露时间的延长,心率下降不明显。结论氯化两面针碱对斑马鱼胚胎有心脏毒性。Objective To study effects of different concentrations of nitidine chloride on the heart development of zebrafish embryos.Methods Zebrafish embryos of 48 hours post-fertilization(48 hpf)were treated with nitidine chloride at various concentrations including 5 mg/L,3.15 mg/L,2 mg/L,1.58 mg/L,1.12 mg/L.The effects of nitidine chloride on the heart development at 60 hpf and 72 hpf were assessed under the microscope.Results Different concentrations of nitidine chloride could cause cardiotoxicity at 60 hpf and 72 hpf.Those embryos mainly showed cardiac arrest and heart hemorrhage when treated with 5 mg/L,3.15 mg/L nitidine chloride,and caused the cardiac malformations when treated with 2 mg/L,1.58 mg/L,1.12 mg/L nitidine chloride.The heart rate of zebrafish embryos increased gradually with the increase of nitidine chloride concentration,but the cardiotoxicity did not increase significantly as the exposure time increased.Conclusion Nitidine chloride has cardiotoxicity on the heart development of zebrafish embryos.
