摘 要:目的:观察针刺对CCS大鼠交感颈中心神经电活动的影响,探讨CCS发病的交感通路机制以及针刺对其干预的作用机制。方法:选取健康Wistar大鼠60只,随机选择其中10只作为假手术对照组,其余用于大鼠CCS复制,然后根据模型成功的评判标准,随机选取模型复制成功的大鼠20只,再分为模型组和电针组,每组各10只。待电针组大鼠清醒后将其放入固定器中,75%乙醇无菌操作人中、风府、内关、心俞,选用1寸毫针,直刺46mm,并连至PCE-A型程控电针治疗仪,电压为5V,频率为2Hz,每次20min,每天1次,连续3天。假手术对照组及模型组大鼠均不予针刺,每天抓空1次。于实验72h后采用Biopac生物信号采集系统记录3组大鼠交感颈中心神经电活动。结果:模型组72h后,大鼠交感颈中心神经放电频率与假手术对照组比较,差异有显著性(P〈0.01);电针组与假手术对照组比较,差异有显著性(P〈0.05);电针组与模型组比较,差异有显著性(P〈0.05)。结论:交感神经系统兴奋性降低可能参与CCS的发生发展,给予针刺早期干预(防治)可调节心交感神经兴奋性,拮抗发生CCS时大鼠心交感神经兴奋性的降低,这可能是针刺防治CCS的作用机制之一。Objective:To observe the effect of acupuncture intervention on electrical activity of cervical sympathetic nerve center in cerebral-cardiac syndrome(CCS)rats,and investigate the mechanism of acupuncture in the prevention and treatment of CCS based on sympathetic pathway.Methods:Sixty Wistar rats of either sex were randomly selected and ten rats were randomly choosed into the sham operation group.Others were intravenous injected with collagenase combined with heparin in caudate nucleus to induce CCS models.According to the standard of successful model,20 rats of successfully induced model were randomly choosed and divided into the model group and the electroacupuncture(EA)group.Each group had 10 rats.The rats in the sham operation group were treated with 0.9% sodium chloride only as control.While the rats in the EA group were put in a fixator,and the points renzhong(DU26),fengfu(DU26),neiguan(PC6),xinshu(BL15)were disinfected by 75% ethanol.Points mentioned above were pierced into 46mm by 1 inch needle,which connected to the PCE-A-type program-controlled electro-acupuncture instrument with a voltage of 5 volt,2 Hz for 20 min a time,1 time per day.Rats in the sham operation group and model group didn’t receive acupuncture.After 72 hours,Biopac biological signal acquisition system was applied to record electrical activity of cervical sympathetic nerve center of rats in three groups.Results:Compared to the sham operation group,the discharge frequency in the model group after 72 hours showed statistic difference(P〈0.01),so was the EA group(P〈0.05);respectively.Compared to the model group,the discharge frequency in the EA group had significant difference(P〈0.05).Conclusion:The reduction of excitability in the sympathetic nervous system might be involved in the process of CCS development.Early acupuncture intervention can adjust the excitability of cardiac sympathetic nerve and resist the decrease of excitability of cardiac sympathetic nerve in CCS rats,which may be on
