摘 要:目的初步探讨活菌卡介苗(BCG)对哮喘小鼠IL-17的调节作用。方法 4周龄雌性Balb/c小鼠30只随机分为A组(对照组)、B组(哮喘组)和C组(BCG干预组),每组各10只。B、C两组予OVA腹腔注射致敏、OVA雾化诱发哮喘,C组在致敏前14d接种BCG。HE染色观察小鼠肺部病理改变,计数支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中细胞总数并分类,酶联免疫吸附试验(ELISA)检测血清及BALF中白介素-17(IL-17)含量。结果肺组织病理观察显示A组气管周围基本无炎症细胞浸润,B组支气管周围大量炎症细胞浸润,杯状细胞增生,C组炎症较B组减轻。B、C组BALF中细胞总数和嗜酸性粒细胞、淋巴细胞百分率均显著高于对照组(P<0.01),而C组嗜酸性粒细胞比例则明显低于B组(P<0.01)。B、C组小鼠血清、BALF中IL-17含量高于A组(P<0.01),而C组则明显低于B组(P<0.01)。各组小鼠血清及BALF中IL-17水平与BALF中嗜酸性粒细胞呈正相关(P<0.01)。结论 BCG接种可抑制IL-17的产生,减轻哮喘小鼠气道炎症反应。Objective To investigate the effect of live mycobacterium bovis Bacille Calmette-Guérin vaccination on the regulation of IL-17 in mice asthma model.Methods 30 female BALB/c mice were randomly divided into 3 groups:group A(control group),group B(asthma group) and group C(BCG group).The mice of group B and C were sensitized by OVA and then stimulated with nebulized OVA,and live BCG were inoculated 14 days before sensitized in group C.The airway inflammation was evaluated by HE staining.Inflammatory cell numbers and morphological identification of leucocytes in bronchoalveolar lavage fluid (BALF) were measured by light microscopy.The levels of interleukin-17 in mice bronchoalveolar lavage fluid (BALF)and serum were measured by enzyme-linked immunosorbent assay (ELISA).Results The neutrophil and eosinophil infiltration in pulmonary tissue was dramatically increased in group B,but no such change in group A and was lighten in group C.The total cell number and the percentages of eosinophils and lymphocytes in BALF of the mice in group B and C were significantly higher than that of group A(P0.01) ,and the percentages of eosinophils in the mice of group C was decreased when compared with that of group B(P0.01).The level of IL-17 in BALF was significantly higher in group B and C than group A(P0.01).In the asthmatic mice,IL-17 expression in the serum and BALF was positively correlated with the percentages of eosinophils in the BALF(P0.01).Conclusion Live BCG vaccination can effectively prevent the development of airway inflammation associated with altered IL-17 levels.
