摘  要:对即墨市大桥盐场的底泥样本进行微生物的可培养研究,分离出1株革兰氏阴性的中度嗜盐菌YC-SA28T。16S rRNA序列比对及系统发育分析结果表明,YCSA28T可能为γ-变形菌纲盐单胞菌属的新种,通过系统分类学研究发现,YCSA28T的16S rRNA序列与亲缘关系最近的盐单胞菌属模式菌株Halomonas ventosaeAl12T同源性为96.9%,其GenBank注册号为FJ984862。YCSA28T的荷兰细菌保藏中心受理序号为NCCB100305T,中国普通微生物保藏管理中心受理序号为CGMCC 1.9150T。YCSA28菌落为乳黄色,细胞杆状或短杆状,周生鞭毛。生长温度范围为5~40℃,生长pH值范围为6.0~9.0,生长绝对盐度(NaCl)范围为2%~15%。氧化酶/过氧化氢酶阳性。辅酶Q-9为主要的呼吸醌,主要脂肪酸为C18:1ω7c(42.88%),C16:0(23.05%)及C16:1ω7c/ω6c(18.00%)。菌株YCSA28T染色体DNA的x(G+C)为63.7%~63.9%,与模式菌株Halomonas vento-saeAl12T间的DNA-DNA杂交率为(45±3)%。结合各种形态及生理生化特征可知,YCSA28T应为盐单胞菌属一新种,建议命名为大桥盐单胞菌(Halomonas daqiaonensissp.nov.)。A moderately halophilic Gram-negative bacteria YCSA28 is isolated from a sedimentary sample at Daqiao saltern in Jimo to study the microorganism cultivation.It is learnt from the 16S rRNA sequence comparison and the systematic growth analysis that YCSA28 can be a noval species of the Halomonas in the Gammaproteobacteria class.As it is shown in its taxological study,YCSA28 has the similarity about 96.9% to its nearest neighour of Halomonas ventosae A112T.A GenBank accession number,FJ984862,is given to the strain.It has been already received and numbered not only as NCCB100305T by NCCB but also as CGMCC 1.9150T by CGMCC.Its colonies appear in cream-yellow.It is in a rod or short rod shape,and with lateral flagella.Growth temperature is between 5~40℃,pH value within 6~9,and growth salinity(NaCl) within 2%~15 %.It is oxidase/catalase-positive.The major quinone of Strain YCSA28 is Q-9,and the cellular fatty acids mainly consist of C18:1ω7c(42.88%),C16:0(23.05%) and C16:1ω7c/ω6c(18.00%).The x(G+C) content of the chromosomal DNA is 63.7%-63.9%.DNA-DNA hybridization between the YCSA28 strain and H.ventosae Al12T has 45±3 % relatedness.It is learnt from the morphological features combined with the physiological and biochemical features that Strain YCSA28 represents a novel species of the genus Halomonas.The name Halomonas daqiaonensis sp.nov.is proposed for Strain YCSA28T.
