摘  要:为了对1株自中草药中分离得到的菌落高度黏连的高产细菌素枯草芽孢杆菌进行有效的活菌计数,试验采用普通平板稀释法进行活菌计数,并对生理盐水、灭菌水、各种PBS、空白培养基等各种不同的稀释液进行比较研究。结果表明:生理盐水、灭菌水、各种PBS均无法将菌团分散,难以形成有效的活菌计数梯度,只有以该菌株的空白培养基作为稀释液时,才能使菌体分散均匀,稀释梯度明显,计数结果准确,重复性好。说明在活菌计数中,空白培养基可能是某些特殊菌株(如菌落高度黏连)的理想稀释液。A strain of Bacillus subtilis with highly sticky colony and high production of bacteriocin was screened from Chinese herb.To get the plate count,physiological saline,sterilized water,phosphate buffered saline(PBS) and sterilized culture medium of this bacterium were used to make different dilutions.The result indicated that the zoogloeas couldn't be separated completely to form valid the counting gradient when using common diluents as physiological saline,sterilized water and PBS while the germs could distribute evenly in its culture medium environment.The accurate plate count with clear gradient dilution grade and stable repeatability suggests that sterile culture medium may be a sort of ideal diluents for some special bacteria such as sticky colony bacterium.
