摘  要:目的:观察腺苷预处理对心肌缺血再灌注损伤的影响,探讨血红素加氧酶-1(HO-1)在其中的作用。方法:雄性SD大鼠18只,体重(250±16)g,随机分为3组,每组6只:①对照组(C组),②缺血再灌注组(IR组),③腺苷预处理+缺血再灌注组(AP组)。采用结扎/放松左冠状动脉前降支建立心肌缺血再灌注损伤模型。再灌注结束后,采用免疫组化法检测心肌HO-1变化,检测心肌梗死面积、心肌组织丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,及血清TNF-α水平。结果:与C组比较,IR组心肌组织MDA含量和血清TNF-α水平升高,SOD活性降低,心肌梗死面积增加(P<0.05),HO-1表达差异无统计学意义(P>0.05);与IR组比较,AP组心肌组织MDA含量和血清TNF-α水平降低,SOD活性增强,心肌梗死面积减少,HO-1表达增加(P<0.05)。结论:腺苷预处理对心肌缺血再灌注损伤的保护作用与HO-1的表达增加有关。Objective: To investigate the role of heme oxygenase-1(HO-1) in adenosine preconditioning against myocardial ischemia/reperfusion injury in rats.Methods: Eighteen male SD rats were randomly divided into three groups(n=6): control group(group C),ischemia/reperfusion group(group IR),and adenosine preconditioning+ischemia/reperfusion group(group AP).The left anterior descending coronary artery was ligated for 30 min and loosed for 120 min to establish the model of myocardial ischemia/reperfusion injury.At the end of 120 min reperfusion,the expression of HO-1 in myocardium was measured by immunohistochemistry.The infarct area,MDA content and SOD activity in myocardium and serum TNF-α concentration were determined.Results: Compared with those in group C,the infarct area,myocardial MDA content,and serum TNF-α concentration were increased,SOD activity was decreased(P0.05),but HO-1 expression had no changes(P0.05) in group IR.Compared with those in group IR,the infarct area,myocardial MDA content and serum TNF-α concentration were decreased(P0.05),along with increased SOD activity and HO-1 expression(P0.05) in group AP.Conclusion: Adenosine preconditioning protects myocardium against ischemia/reperfusion injury,and up-regulation of HO-1 expression may be involved in this protecting process.
