摘 要:目的:探讨金丝桃素单体、苦味叶下珠水溶液浸出物、茵陈水溶性提取物体外抗人巨细胞病毒(HCMVAD169)的作用。方法:采用细胞病变(CPE)抑制法观察金丝桃素单体、苦味叶下珠水溶液浸出物、茵陈水溶性提取物对HCMV AD169毒株的抑制作用,使用MTT比色法检测细胞的损伤程度,并利用吸光度(A)值评价这三种中药抗HCMV的效应。结果:金丝桃素单体、苦味叶下珠水溶液浸出物、茵陈水溶性提取物半数中毒浓度(TC50)分别为195.96,310.57,904.49 mg/L;半数有效浓度(IC50)分别为19.15,51.25,195.11 mg/L;治疗指数(TI)分别为10.23,6.06,4.64;IC50下,对HCMV的抑制率分别为47.6%,41.7%,42%。细胞培养96 h,IC50下,光学倒置相差显微镜下:金丝桃素单体抑制组中约30%贴壁细胞变成圆型或发生肿胀,而另外两种中药抑制组中约45%-55%贴壁细胞变成圆型或发生肿胀。金丝桃素单体、苦味叶下珠水溶液浸出物、茵陈水溶性提取物溶液分别在6.25,10,3.2 mg/L浓度下具有显著的抗HCMV的作用,且随药物浓度的增高其作用而增强。苦味叶下珠水溶液浸出物和茵陈水溶性提取物抗HCMV作用的差异无显著性(P>0.05),而金丝桃素单体与其他两种中药抗HCMV作用的差异有显著性(P<0.05)。结论:金丝桃素单体、苦味叶下珠水溶液浸出物及茵陈水溶性提取物均是理想的抗HC-MV中药,其中金丝桃素单体抗HCMV的作用尤为突出。Objective: To study the difference of inhibitory effect of monomeric hypericin,lixivium of Phyllanthus Uyinayia L solution and distillable matters with water-solubility from Artemisia Capillaris Thunb on human cytomegalovirus(HCMV) in vitro.Methods: Cytopathic effect(CPE) method was used to examine the inhibitory effect of the three botanic medicines on HCMV AD169.MTT colorimetric assay was used to test injury degree of cells.The inhibitory effect of the three herbs was evaluated by measuring the values of absorbance(A).Results: The median toxic concentration(TC50) was respectively 195.96,310.57,and 904.49 mg/L,median effective concentration(IC50) was respectively 19.15,51.25,and 195.11 mg/L,therapeutic index(TI) was respectively 10.23,6.06,4.64,and inhibitory ratio was respectively 47.6%,41.7%,and 42% for the three herbs.At the concentration of IC50,after 96 hours,inverted phase contrast microscope showed 30% cells along wall were round or swollen in monomeric hypericin group,while this ratio ranged between 45% to 55% in the other two herbs groups.The obvious inhibitory effect of the three herbs at the concentration of 6.25,10 and 3.2 mg/L,respectively,were significant.It suggested that the three herbs could inhibit HCMV in a dose-dependent maner.Inhibitory effect between the lixivium of solution Phyllanthus Uyinayia L and distillable matters with water-solubility from Artemisia Capillaris Thunb on HCMV were not different in vitro,but inhibitory effect of monomeric Hypericin on HCMV in vitro were better than those of the others.Conclusion: All the three herbs are ideal traditional Chinese medicines in inhibiting HCMV in vitro,especially monomeric hypericin.
