摘 要:利用微生物发酵制备γ-聚谷氨酸,降解得小分子γ-聚谷氨酸,将天冬氨酸接入小分子γ-聚谷氨酸,得γ-聚谷氨酸-天冬氨酸聚合物(GAP460)。核磁共振、红外光谱、差热扫描量热分析(DSC)等方法表征GAP460;常规方法检测GAP460的结构稳定性,免疫原性,急性毒性及血液参数值等。结果表明,成功制备了γ-聚谷氨酸-天冬氨酸聚合物,其在中性、常温及4℃条件下结构稳定;GAP460免疫小鼠,初次免疫后八周测得抗体滴度为1∶20,显示GAP460是低免疫原性;急性毒性、血液参数等生物相容性指标显示该材料无毒。Poly(γ-glutamic acids) was obtained by using fermentation methods.The small molecular weight of poly(γ-glutamic acids) was prepared by acid hydrolysis.The aspartic acid as a side chain was linked to poly(γ-glutamic acids),and poly(γ-glutamic acids)-aspartic acid(GAP460) was prepared.The polymer was characterized by 1H-NMR,X-ray and differential scanning calorimetry.Structural stability,immunogenicity,acute toxicity and principal haematological parameters of GAP460 were examined by routine methods.Results reveal GAP460 is obtained and structural of GAP460 is stable in the conditions of neutral,room temperature and 4 ℃,on eight weeks after first immunization,the serum antibody titer of rats is 1∶20,GAP460 shows good safety and low immunogenicity in rats,acute toxicity and principal haematological parameters studies show GAP460 displays atoxic.Our results suggest that GAP460 can be used as an effective carrier for drug delivery and may have potential applications.
