IL-10与IL-12在银屑病患者系统血液快速固有免疫反应中的表达

摘　　要：目的：检测银屑病患者IL-10及IL-12在体外系统血液快速固有免疫反应体系中的表达。方法：将灭活大肠杆菌（3X108／mL）0．2mL做为免疫原加入抗凝全血细胞悬液0．2mL和血浆0．3mL中，37℃水浴1h，离心，取上层血浆，固相夹心法酶联免疫吸附试验（ELISA）测定血浆IL-10及IL-12含量。结果：银屑病血液经免疫原激活后IL-12含量明显上升，IL-10含量无明显上升，但正常人免疫原激活后IL-12和IL-10含量都明显上升。银屑病血液经免疫原激活后IL-12／IL-10比值增高。结论：银屑病患者血浆中IL-12与IL-10细胞因子水平的异常是固有免疫反应亢进的一种表现，可能与银屑病的系统免疫学发病机制相关。Objective： To detect the expression of IL - 10 and IL - 12 in patients with psoriasis by systematic hematogenic immunoreaction. Method： Escherichia coli suspension （3×108/mL） 0.2 mL was added to anticoagulate whole blood with citric acid and 0.3 mL plasma, followed by lh incubation at 37℃. The expression of IL- 10 and IL- 12 in plasma was determined by ELISA. Results： The IL- 12 level in psoriasis patients was significantly increased after immunogen activation, while the IL- 10 level was not significantly increased. In healthy subjects, both levels of IL - 10 and IL - 12 were increased significantly after immunogen activation. The plasma IL - 12/IL - 10 ra- tio increased after immunogen activation in psoriasis patients. Conclusion： The abnormal levels of IL- 10 and IL- 12 show the over- expression of innate immune in psoriasis patients, which may play an important role in systematic immune pathogenesis of psoriasis.
