摘 要:目的 :了解蓝藻提取物对原代培养大鼠肝细胞和人类肝细胞系的毒作用 ,为有效防治这类毒素中毒及明确这两类细胞在体外试验中的价值提供依据。方法 :用蓝藻提取物处理上述两类细胞后 ,观察它们的形态变化及测定乳酸脱氢酶渗出率。结果 :在蓝藻提取物作用下 ,原代大鼠肝细胞形态发生一系列变化 ,且乳酸脱氢酶渗出率随提取物剂量和处理时间的增加而增加 ;人类肝细胞系上述两项指标都没有明显改变。结论 :蓝藻提取物对两类肝细胞的毒作用有明显差别 ,这可能与两类细胞上胆酸转运系统的活力不同有关。研究蓝藻对人类肝脏的危害尚需寻找其他更敏感的体外实验对象。Objective:The research was made to understand the toxic effect of cyanobacteria extract (CE) to primary cultured rat hepatocytes (PCRH) and Chang liver cell (CLC) because the cyanobactenria pollution in water body has been a serious health problem around the world Methods:After two kinds of cells were treated by CE,the morphological changes of cells were observed with microscopy and LDH leakage was detected Results:Morphological changes were easily detected and LDH leakage increased in a dose and time dependent manner in PCRH But neither morphological changes nor LDH leakage increase was observed in CLC Conclusions:There was a significant difference between two kinds of cells about toxic effect of CE More sensitive cells have to be found for research on damage of miorocystins to human liver
